STATEMENT OF WORK
Hi-C Sequencing and Proxi-Meta Assembly of Metagenomes from (9) Pig Fecal Samples.
Objective:
The Contractor will provide Hi-C library preparation and sequencing and bioinformatic assembly for nine pig fecal samples collected by Agriculture Research Service, Animal Biosciences and Biotechnology Laboratory (ABBL) researchers. Sequence data will be delivered to researchers within ARS-ABBL.
Background
US Dept of Agriculture, Agricultural Research Service (ARS), Animal Biosciences and Biotechnology Laboratory (ABBL) requests a Contractor provide sequencing services which will produce a set of nine Proxi-Meta Hi-C sequencing datasets from nine pig fecal samples.
In a typical microbiome sequencing project, DNA is extracted from diverse microbial cells in a single sample, sequenced, and then bioinformatically assembled and assigned to individual species. These large and complex datasets require specialized bioinformatic workflows which are often error prone.
To circumvent these errors and produce a more reliable set of final microbial genome assemblies, Proxi-Meta Hi-C technology begins with a laboratory procedure which links all the DNA within a single cell before DNA extraction and sequencing. Then, a specialized collection of bioinformatic tools is used to produce a final, accurate, set of genome assemblies for multiple microbial species.
This service will thus facilitate the acquisition of accurate microbial genomes for microbes present in nine pig fecal samples. These high quality microbial reference genomes will facilitate the advancement of multiple projects proposed to meet National Program objectives.
