About this file

This is a Statement of Work for a NextSeq1000 to NextSeq2000 upgrade service to be performed by Illumina for the National Institutes of Health (NIH), National Eye Institute (NEI). The objective is to upgrade the government's current NextSeq1000 sequencing system to NextSeq2000 capability, which will expand the system's sequencing flexibility by enabling use of all four flow cell types (P1, P2, P3, and P4) rather than the P1 and P2 limitations of the 1000 model. The upgraded system will support sequencing capacities ranging from 100 million to 1.8 billion reads depending on flow cell selection.

The contract is structured as a fixed-price engagement with an estimated performance period of one week. Work will be performed on-site at NIH, Building 6, Room 328. The contractor (Illumina) is responsible for supplying all parts, equipment, and materials required to complete the upgrade, with no Government Furnished Equipment or Information to be provided by NEI. The primary deliverables are all supplies necessary for the upgrade and a fully functional NextSeq2000 instrument. The government will coordinate scheduling with the contractor to identify an appropriate time for the work. No security requirements are associated with this engagement.

Statement of Work (Illumina)-2025.

1. Delivery Order Title and Scope:

NextSeq1000 to NextSeq2000 upgrade.

This work is intended to convert our current NextSeq1000 sequencing system to a NextSeq2000

2. Background:

The NextSeq 1000 and NextSeq 2000 Sequencing systems are bench top platforms that utilize innovative patterned flow cell technology for diverse sequencing applications. They utilize XLEAP-SBS chemistry, a faster, higher quality, and more robust SBS chemistry built on the proven foundation of standard Illumina SBS chemistry. XLEAP- SBS nucleotides use state-of-the-art dyes and novel linkers and blocks that are more resistant to heat, show a 50× reduction in hydrolysis, and 2.5× faster block cleavage to reduce phasing and prephasing. The XLEAP-SBS polymerase is engineered to incorporate nucleotides faster and with higher fidelity than ever before. There are four different flow cells with varying sequencing capacity that allow for maximum sequencing flexibility.

The P1 flow cell can produce up to 100 million paired reads while the P2 flow cells can generate 400 million reads. The high capacity P3 flow cell can produce up to 1.2 billion reads and the P4 flow cells up to 1.8 billion reads.

In terms of the differences between the 1000 and 2000 systems, the NextSeq 1000 can only use the P1 and P2 flow cells while the NextSeq 2000 sequencer can use all four flowcells. Thus, you can have maximum flexibility with the NextSeq 2000 instrument.

3. Objectives:

a. The goal of this project is to upgrade our NextSeq 1000 machine to a NextSeq2000.

4. Requirements:
