STATEMENT OF WORK

NO:

I. GENERAL INFORMATION

A. INTRODUCTION

The U.S. Geological Survey has multiple molecular-based projects focused on taxonomy and gene flow of various vertebrate species. The project will require sample preparation, sequencing, and preliminary bioinformatics analysis of DNA sequences of various vertebrate species. Due to the uncertainty of the number of samples that will need to be sequenced each year, the request for number of samples is not able to be defined. In order to have the flexibility to process samples as they are available (be they more or less each year), we would like to set up a BPA for five years with no yearly minimum or maximum number of samples or orders, but an NTE (not to exceed) amount of funds to use over 5 years totaling $109,000.00. No funds will be obligated at the time of BPA setup. All requests for analysis / services will be processed from separate Purchase Requests and obligated funds through separate BPA calls.

B. BACKGROUND

Scientists (Nathan Johnson and Margaret Hunter) at USGS-Wetland and Aquatic Research Center (WARC) have already optimized protocols and begun processing samples for this project using Diversity Arrays Technologies (DArT). Now that the PIs are funded and will be managing multiple projects requiring services from DArT, we are submitting a DI-1 to continue having samples processes by DArT to continue these projects and ensure consistency with data collection and promised deliverables to our funders.

Definitions:

DArT = Diversity Arrays Technologies (DArT): the company with the patented technology for genotyping by hybridization

Genotyping by hybridization: patented method that allows relatedness to be measured between individual organisms without the need for a map of the organism’s genome.

C. SCOPE

The contractor shall furnish all labor, supervision, materials, supplies, tools, equipment, and expertise necessary to perform the genotyping by hybridization of the samples. The contractor shall work in coordination with the client agency.

The period of performance will be for (1) year from date of award with four (4) option years (five total years) for additional sample processing to ensure consistency of sequence data throughout the duration of the projects. The USGS is conducting a series of large-scale molecular projects to investigate the genetic diversity and distributions of imperiled and invasive species in North America. The objectives are to determine the relationships between individuals from different populations to better help our partners conserve imperiled and control invasive species. During the period of performance, is anticipated that four plates of samples will be sent for the services during the base year.

II. WORK REQUIREMENTS

A. TECHNICAL REQUIREMENTS

The contractor shall be responsible for conducting genotyping by hybridization (GxH) for samples submitted by USGS and make a description of methods and resulting files (.xlsx files) accessible to the USGS (via internet, LIMS, or email).

The Contractor is responsible for receiving the samples and all molecular processing of the samples therein.

Responsibility and liability for loss or damage of samples shall remain with the contractor until final inspection and acceptance of sequences by the USGS.

B. ACCEPTANCE CRITERIA

Resulting files (e.g., .xls and fastq/GZ) will be made accessible to USGS (via internet) along with a description of methods used to generate the data and results. All samples must be processed in a timely manner. Most samples should be processed within one or two months of submission, except for very busy times or when large numbers of samples are submitted simultaneously.

C. DELIVERABLES

All sequence data shall be delivered in a secure manner and be of publication quality.

III. SUPPORTING INFORMATION

A. Place of Performance: Contractor’s facility
B.Period of Performance: Date of Award to five years. (BPA)
C.Government Furnished Property (GFP): N/A
D.SPECIAL CONSIDERATIONS.

1. Key personnel qualifications Laboratory must be operated and supervised by a staff member/project manager with expertise in genotyping by hybridization, sequencing and analysis techniques.

2. Data Rights, Safety, Environment.

a. Responsibility and liability for loss or damage of samples shall remain with the contractor until final inspection and acceptance of sequences by the USGS. All sequences shall be delivered in a secure manner and be of publication quality.

b. The Contractor is responsible for offsite disposal of all chemical waste generated during the sample preparation and sequencing process.
