SPECIFICATIONS
Seed microbiome sequencing service
1. Introduction: Scientists at the Genomics and Bioinformatics Research Service, Stoneville MS.
Has a need to microbiome analysis using plant tissues as the starting material.
Scope: The service requirement is for a microbiome core facility capable of extracting bacterial and fungal DNA from plant seed tissue, preparing libraries for amplicon and next generation metagenomic sequencing, and sequencing those samples for fungal and bacterial taxa. Specifically, the requirement has to produce quality microbial DNA extracted given the high prevalence of plant DNA present in each sample. The DNA extracted from samples designated for amplicon sequencing must be able to be aliquoted for both 16S rRNA and ITS sampling, that is, these sequencing types must occur on material from the same sample.
Technical Requirements: Said service must meet the requirement to produce quality microbial DNA extracted given the high prevalence of plant DNA present in each sample. This is in part achieved by providing a validation run of four samples
The Contractor will perform 1) DNA extraction from plant material; 2) library preparation; 3) amplicon and whole genome DNA sequencing of bacterial communities; 4) amplicon sequencing of fungal communities; and 5) performs microbial sequencing from plant material; 6) provides a validation run of four samples
