Statement of Work Arctic Cod RNA sequencing
NFFP7210-20-02784
1.0 Organization
The Northwest Fisheries Science Center (NWFSC), National Marine Fisheries Service (NMFS), National Oceanic & Atmospheric Administration (NOAA), Department of Commerce (DOC) is a government agency charged with the mission of stewardship for living marine resources.
2.0 Background and Objective
This project is part of a larger ongoing study aimed at understanding the long-term impacts of oil spills on spawning populations of fish, with a special emphasis on forage fish populations of high latitude regions. Previous work on the acute embryonic effects of oil exposure in Arctic Cod demonstrated toxicity to the developing heart, also leading to craniofacial defects and defects in lipid metabolism in hatched larvae. In the current studies, the morphological and physiological effects were measured in embryos during and immediately after acute acute oil exposure, and in newly hatched larvae after completing incubation in clean water. These effects were measured in three separate experiments with two different oil sources (Alaskan and Norwegian). Samples were collected that allow comparisons of the same exposure doses duplicated across years and for the two different oil types. The objective of the current SOW is to identify the changes in gene expression associated with these measured effects. Individual cDNA libraries for each sample will be prepared by the NWFSC and combined into 15 pooled sets for RNA sequencing.
3.0 Services Required
This contract entails a single task: multiplexed mRNA sequencing of Arctic cod (Boreogadus saida). Sequencing of 124 samples using NovaSeq PE150 Illumina technology with mRNA library preparation and poly A tail enrichment. Sequencing should produce greater than 12 million paired end 150-nucleotide reads per sample (greater than 4 Gb of data per sample) and the vendor should provide monitoring results for the data quality control. Sequencing data is required to be delivered 40 business days after sample submission.
4.0 Deliverables
Sequence data delivered electronically (via download link) in .fasta format.
5.0 Period of Performance:
August 1, 2020 through November 30, 2020
