About this file

This document is a Statement of Work (SOW) for a federal contract opportunity issued by the National Institutes of Health (NIH) National Institute on Aging. The SOW is for Next Generation Sequencing Analysis of 160 human post-mortem frontal cortex tissue samples to identify potential therapeutic targets and biomarkers for early-onset and late-onset Alzheimer's disease. The contractor shall provide the sequencing services, including library preparation, sequencing on the NovaSeq X Plus platform, and delivery of raw FASTQ data files. The period of performance is 12 months from date of award. The associated federal contract opportunity, Solicitation Number 75N95024Q00353, is a full and open commercial item acquisition with no set-asides, valued under the simplified acquisition threshold. Responses are due by the closing date and must include pricing, technical capability, and past performance information. Award will be made on a firm-fixed-price basis to the offeror whose proposal is most advantageous to the government, with technical and past performance factors considered significantly more important than price.

FAR Part 11—Describing Agency Needs

STATEMENT OF WORK

GENERAL INFORMATION

Next Gen Sequencing Analysis in Post-Mortem Frontal Cortex Tissues

The NIH Intramural Center for Alzheimer’s Disease and Related Dementias is an ambitious program to genotype disease patients and controls around the world to further understand the genetic architecture of Alzheimer’s Disease and related dementias. There is much to learn about genetic risk factors and the path to further understanding requires working collaboratively and openly sharing data, processes, and results.

Understanding the molecular signature of human brain is essential to study pathogenesis of neurodegenerative diseases. High-throughput Next Generation Sequencing will assist in epigenetic analysis in post-mortem frontal cortex tissue. Our goal is to identify potential therapeutical targets and biomarkers in patients with early-onsite and late-onsite Alzheimer’s disease.

SCOPE OF WORK

Independently and not as an agent of the Government, the Contractor shall furnish all the necessary services, qualified personnel, material, equipment, and facilities, not otherwise provided by the Government as needed to perform the Statement of Work below.

Next Generation sequencing of 160 human samples. Next Generation Sequencing services must include:

• Application: proteomics (run only)

• Species: human

• Starting material: premade Illumina-compatible libraries prepped with Alamar NULISA kit

• Sample size: 160

• Sequencing platform: NovaSeq X Plus 10B (100cycle), 20 flow cells

• Output: ~1.25B reads pair from a lane

• Deliverables: raw data (fastq) only

• Data delivery method: basespace transfer

• Turnaround time: 2-3 weeks after a batch of 8 libraries pass their initial QC

• A 100 cycle kit must be used to conduct a 34-cycle single-end run.

• Vendor must deliver fastq data and the file will then be uploaded by the Government to the ARGO

Command Center to obtain an NPQ count file.

• Vendor must show evidence of Good Laboratory Practices (GLP) and must provide at least two (2)

General Requirements:

Specific Requirements:

of the certifications and/or accreditations shown below:

• Vendor must be ISO 9001 certified based on quality management principles.

• Vendor must be cGLP certified which ensure current good laboratory practice regulations as defined by the FDA.

• Vendor must be CLIA certified which ensures Clinical Laboratory Improvement Amendments of

1988 are practiced.

• Vendor must maintain a CAP laboratory accreditation to ensure precise testing results.

LEVEL OF EFFORT

Vender must provide Next Generation Sequencing information by fastq raw data files. Files must be reported thru Google Cloud Server, and communication regarding results must be conducted via email.

GOVERNMENT RESPONSIBILITIES

Premade Illumina-compatible libraries prepped with Alamar NULISA kits will be submitted to the contractor as the starting material for which to process and sequence.

DELIVERY OR DELIVERABLES

Raw data in FASTQ format delivered by Google Cloud Server.

REPORTING REQUIREMENTS

The contractor must deliver fastq data and the data files will then be uploaded by the Government to the ARGO Command Center to obtain an NPQ count file.

TRAVEL

The principal place of performance is the contractor’s work site. No travel is necessary for this project nor will any travel expense be reimbursed.

KEY PERSONNEL

Contractor shall provide a dedicated project manager to the government as a contact point for the services.

PUBLICATIONS AND PUBLICITY

Contractor shall not, nor is required to, publish any articles of data as a result of fulfilling this requirement.

Other Considerations:

CONFIDENTIALITY OF INFORMATION

Government will not be providing Contractor with confidential information of a personal nature about individuals. All data submitted by the Government to the Contractor for processing shall be deidentified.

DATA RIGHTS

The National Institute on Aging shall have unlimited rights to and ownership of all deliverables provided under this contract. Contractor will not release or share data without government approval. Contractor will not use data for scientific investigation and will have no right to work with these data or publish using these data. FAR clause 52.227-14, Rights in Data—General, shall be applicable to any resulting contract.

SECTION 508—ELECTRONIC AND INFORMATION TECHNOLOGY STANDARDS

The contractor shall comply with Section 508 of the Rehabilitation Act (29 U.S.C. § 794d), as amended by the Workforce Investment Act of 1998 (P.L. 105-220), August 7, 1998.
