STATEMENT OF WORK
GENERAL INFORMATION
Title of Project:
Neurodegenerative disease research
Statement of Need and Purpose:
To enable us to examine alternative splicing and transcript missplicing in particpiants with Alzheimer’s disease and related dementias, we will use long read RNASeq. We will use single cell 10x Genomics libraries on the PacBio long read sequencing platform. Long read sequencing from single cells are necessary for us to be successful in completing our research project’s aims.
Only the PacBio sequencing platform is capable of sequencing the SMRTbell libraries created from the Kinex kit.
Background Information and Objective:
Understanding the molecular signature of human brain is essential to study pathogenesis of neurodegenerative diseases. High-throughput long read RNA sequencing will assist in epigenetic analysis in post-mortem frontal cortex tissue. Our goal is to identify potential therapeutical targets and biomarkers in patients with early-onsite and late-onsite Alzheimer’s disease.
Period of Performance:
6 months from date of award.
SCOPE OF WORK
General Requirements:
Independently and not as an agent of the Government, the Contractor shall furnish all the necessary services, qualified personnel, material, equipment, and facilities, not otherwise provided by the Government as needed to perform the Statement of Work below.
Specific Requirements:
• Contractor shall perform HiFi sequencing on Government provided 36 PacBio Kinex libraries using the PacBio Revio platform.
• Contractor shall ensure that an accuracy of 90% of bases >Q30 and median read accuracy of >Q30.
• Contractor shall ensure a density of 25 million zero-mode wavelengths is employed in data collection.
• Contractor shall ensure fundamental processing steps are performed on instrument including barcode demultiplexing and conversion to standard BAM format.

• Contractor shall provide reads that average 1000 bp in length for cDNA from 10x Genomics libraries
• Contractor shall provide 6 million reads per SMRT cell
LEVEL OF EFFORT:
Contractor shall provide the assay results via reporting transfer thru email, along with routine trouble shooting communications.
GOVERNMENT RESPONSIBILITIES
Government will provide 36 PacBio Kinex libraries that are compatible with running on the Revio
DELIVERY OR DELIVERABLES
Raw data delivered by Google Cloud server.
REPORTING REQUIREMENTS
Contractor shall provide documentation regarding:
a. From our single nuclei long read RNASeq libraries
OTHER CONSIDERATIONS
Travel:
The principal place of performance is the Contractor’s work site. Travel for contractor employees shall not be necessary.
Key Personnel:
Contractor shall provide a dedicated project manager to the government as a contact point for the services.
