About this file
This is a Statement of Need for a federal procurement by the National Heart, Lung, and Blood Institute (NHLBI), part of the National Institutes of Health.
The NHLBI's Regenerative Therapies for Inherited Blood Disorders Lab seeks to procure the STEMvision™ Automated CFU Reader system to standardize Colony-Forming Unit assay analysis. Currently, the lab manually identifies and counts colonies using inverted light microscopy, a labor-intensive process requiring specialized expertise that is prone to observer variability and impacts research reproducibility. The required system must provide automated, high-resolution imaging with sophisticated software algorithms to identify and classify hematopoietic colonies (erythroid, myeloid, and mixed colonies) with high accuracy. Key specifications include: automation with "walk-away" imaging and automated counting for 6-well plate formats; processing speed of approximately 5 to 10 minutes per complete plate; pre-validated algorithms for human or mouse hematopoietic assays to eliminate technician bias; optimization for use with meniscus-free cultureware (SmartDish™) and specialized methylcellulose media (MethoCult™); and permanent digital records for retrospective review and audit trail compliance. The procurement includes two line items: one STEMvision Unit with 1-year warranty and 4 analysis packs (Part Number 22010) and one STEMvision Mouse Algorithm Bundle (Part Number 22012). Delivery is required to Fariba F. Chinian, Lab Manager, at the Clinical Research Center in Bethesda, Maryland. The point of contact will be provided upon award.

Statement of Need
1. Project Title STEMvision™ Automated CFU Reader for Regenerative Therapies for Inherited Blood Disorders Lab ( Dr.
Larochelle’s lab) / NHLBI
2. Statement of Need/Purpose and Objectives Regenerative Therapies for Inherited Blood Disorders Lab ( Dr. Larochelle’s ) at NHLBI currently performs Colony-Forming Unit (CFU) assays to assess the proliferation and differentiation potential of hematopoietic progenitor cells. Currently, these colonies are identified and counted manually using inverted light microscopy.
This process is labor-intensive, requires highly specialized expertise, and is inherently prone to intra- and inter-observer variability, which can impact the reproducibility of critical research/clinical data. The purpose of this acquisition is to procure an automated colony-counting instrument—specifically the STEMvision™ system—to standardize CFU assay analysis. The system must provide automated, high-resolution imaging and sophisticated software algorithms to identify and classify hematopoietic colonies (e.g., erythroid, myeloid, and mixed colonies) with high accuracy and speed.
3. Product Specifications/Features/Salient Characteristics of STEMvison Automation: Provides "walk-away" imaging and automated counting of CFU assays in a 6-well plate format.
Speed: Capable of imaging and analyzing a complete 6-well plate in approximately 5 to 10 minutes.
Standardization: Uses pre-validated algorithms for human or mouse hematopoietic assays to eliminate technician bias.
Compatibility: Optimized for use with meniscus-free cultureware (e.g., SmartDish™) and specialized methylcellulose media (e.g., MethoCult™) to ensure image clarity.
Data Integrity: Provides a permanent digital record of the entire culture well for retrospective review and audit trails.
