About this file
This document is a Statement of Need (SON) for the procurement of one (1) Teledyne Princeton Instruments ProEM-HS 512BX3 electron-multiplying charge-coupled device (EMCCD) Camera. The Optical Microscopy Core (OMC) of the Laboratory of Receptor Biology and Gene Expression (LRBGE) at the Center for Cancer Research (CCR), National Cancer Institute (NCI), National Institutes of Health (NIH) requires this camera to support its custom-built highly inclined and laminated optical sheet (HILO)-illumination system used for Single Molecule Tracking (SMT) research on cancer cells.
The government plans to procure the camera on a sole source basis from Teledyne Princeton Instruments, as the ProEM-HS 512BX3 camera is compatible with the existing HILO-illumination system and cannot be replaced by a different brand without rebuilding the entire system. The camera must meet specific technical requirements including size, compatibility with Micro-Manager 2.0 software, pixel size, cooling, read noise, quantum efficiency, and frame rate. Delivery is required within 10 weeks of award, with installation within 14 calendar days. Payment will be made in full upon successful delivery, installation, and acceptance.

STATEMENT OF NEED (SON)
Reference# 75N91024Q00032 1
1.0 TITLE
Procurement of One (1) Teledyne Princeton Instruments ProEM-HS 512BX3 EMCCD Camera
2.0 BACKGROUND
The Optical Microscopy Core (OMC) of the Laboratory of Receptor Biology and Gene Expression (LRBGE) at the Center for Cancer Research (CCR), National Cancer Institute (NCI), National Institutes of Health (NIH), uses a custom-built highly inclined and laminated optical sheet (HILO)-illumination system for Single Molecule Tracking (SMT) for both basic and translational research of cancer cells. SMT is a state-of-the art technique that supplies new insights by providing biophysical information on the protein complex activities at the level of individual molecules.
Specifically, fluorescently tagged transcription factors are analyzed as they move around inside of the cell nucleus sampling chromatin sites to locate their target promoters. Experiments with this custom-built fluorescence microscope system allow a determination of how these molecules diffuse inside of the nucleus and also probe their binding interactions with chromatin. To visualize individual molecules, the OMC/LRBGE labels HaloTag and SNAP-tag fusions with special bright and photostable ligands suitable for the long-term single molecule tracking. The HILO-illumination imaging system is used for the imaging and quantification of transcription factors interacting with DNA in murine and human cancer cells.
To support the custom-built HILO-illumination system, LRBGE Optical Microscopy Core requires one (1) Teledyne Princeton Instruments ProEM-HS 512BX3 electron-multiplying charge-coupled device (EMCCD) Camera.
