STATEMENT OF NEED FOR SUPPLY OR SERVICE
PURPOSE:
The Fisheries Resources Division and the Offshore Wind Monitoring Program (OWMP) of the
Ecosystem Science Division (ESD) of the Southwest Fisheries Science Center, La Jolla, CA has the responsibility to manage finfish populations along the US EEZ in support of the Magnuson Stevens
Fisheries Management Act and to understand the impacts of offshore wind on the pelagic ecosystem. To meet these responsibilities, the SWFSC Center conducts research to determine the origin of fish in the US
EEZ. The analysis of the micro-chemical constituents that are contained in the otoliths of commercially and ecologically important fishes provide a record of growth and the environment (water temperature and salinity) when the fish was born. Such constituents also provide a record of the ocean environments transited during migrations along the west coast of the US. Of particular interest is the Pacific sardine, a small pelagic fish that migrates along the coast from Mexico to Canada and is a species of commercial and ecological importance. Identifying the origin of these fishes using standard chemical approaches applied to material ground from their otoliths is complicated by their smaller size (<5mm) resulting very small amounts of material for conventional stable isotope analysis, and so specialized equipment and expertise is needed to accomplish this task.
Thus, the SWFSC has the need for the processing and analysis of up to 96 sardine otoliths using micro-griding, micromilling and micro-scale (0.2 to 0.6 microgram samples) for stable isotopes. Otolith margins
(last two otolith increments) shall be micromilled using a GEOMILL326 (or equivalent) and stable isotope analysis of the margins should be accomplished using a micro-scale analytical system (MICAL3c) coupled with an IsoPrime100 Mass Spectrometer (or equivalent). Standard QA/QC procedures including blank samples and known standards shall be used between each otolith sample so that sensitivity of the technique can be determined. Otoliths will be provided to the selected Vendor, and data will be provided in electronic media
We expect to award a Firm-fixed price contract, with no option years for delivery by August 01, 2025.
TECHNICAL SPECIFICATIONS:
1) Capacity to micro-mill margins of sardine otoliths, and collect material sufficient for stable isotope chemistry.
2) Micro-scale stable isotope techniques must be able to process as little as 0.2 ug (typically 0.6 micrograms)
3) Analysis of carbon and oxygen stable isotopes along with analysis of blanks to determine precision and accuracy of stable isotope in otoliths
DELIVERABLE SCHEDULE:
All analyses shall be completed by August 01, 2025.
