About this file

This document is a Sole Source Justification template for a federal procurement under the Simplified Acquisition Threshold. The requirement is for DNA extraction and genotyping services for pecan and hickory samples, using a specific 3,100 SNP marker panel developed by Cornell University and Diversity Arrays Technology (DArT). The anticipated period of performance is a base year with two option years. The estimated dollar value is not provided.

The government has determined that DArT is the only vendor capable of meeting this requirement, as the SNP marker panel was jointly developed by DArT and Cornell University. The government intends to award a sole source purchase order to DArT under FAR 13.106-1(b)(1). The government has posted a Sources Sought notice and conducted market research, which are attached. Follow-on work for additional sample testing in future years is expected.

SOLE SOURCE JUSTIFICATION

MANDATORY TEMPLATE FOR FEDERAL PROCUREMENTS UNDER THE SIMPLIFIED ACQUISITION THRESHOLD

1. Estimated Dollar Value:

2. Anticipated Period of Performance: Base year and 2 option years

3. Does the anticipated Period of Performance include option years? If no, explain. Yes

4. Sources Sought Notice posted to Contract Opportunities and attached to this document?

Yes No

5. Market Research conducted and attached to this document? If not, explain. Yes

6. Is this a new requirement or a follow-on to an expiring award?

New requirement Follow-on to an expiring award

7. Describe the requirement. Please note that the requirement is the minimum acceptable specification or capability, and is not necessarily the best in the industry. Please note whether any additional follow-on work is anticipated in response to the current proposed effort.

DNA extraction from 50 “plates” of racked 1.1mL microtubes in a 96-well arrangement (~4,700 samples).

• Genotypes for 3100 SNP positions specified in the open-source “Pecan_DArTag_BI_Cornell_University (1.0)panel with 80bp flanking sequence (40bp on each side of the SNP marker, total 81bp).

• Fastq file delivery for each 81bp microhaplotype detected in each sample and at each locus

• Missing Allele Discovery Counts (MADC) file

• Follow-on work in future years is expected (estimating a minimum of 30 plates sequenced in FY25, andFY26)

8. List the proposed vendor and contact information.

9. Describe why the proposed vendor is the only vendor from which the requirement can be obtained. Examples of acceptable justifications include but are not limited to: sole manufacturer and seller, adherence to warranty provisions, urgency, etc. If other vendors can provide similar products or services, describe why they cannot meet our particular requirement.

Commonly available SNP genotyping services are not targeted and provide random SNP markers sequenced throughout the genome. We require uniform genotype information for all samples at 3100 specific SNP marker locations with 80bp of flanking DNA sequence. A USDA-ARS funded initiative at Cornell University (Breeding Insight), in conjunction with Diversity Arrays Technology (DArT) developed and validated this 3100 SNP marker set using DNA sequencing data provided by our program on the top 48 pecan cultivars used in the breeding program.
