About this file

This document is a Sole Source Justification and Approval for the procurement of a DNA purification system for the USDA Forest Service Pacific Northwest Research Station. The agency requires a Thermo Scientific KingFisher Duo Prime Purification System with SmartStart to support two Bipartisan Infrastructure Law (BIL) projects, ERR09 and ERR10, which involve detecting invasive forest pathogens in the US and internationally. The system is the sole source as it provides unique magnetic bead technology to extract high-quality and high-yield DNA, RNA, and proteins from various biological samples, which is essential for the projects' success. The estimated total value of the procurement is not provided. The agency intends to award a sole source contract to Thermo Fisher for the DNA purification system, as the conventional equipment used previously was unable to produce the required DNA quality and yield from the forest soil samples.

SOLE SOURCE or BRAND NAME JUSTIFICATION AND APPROVAL FOR

PURCHASES NOT EXCEEDING THE SIMPLIFIED ACQUISITION THESHOLD

1. DESCRIPTION OF THE SUPPLIES/SERVICES REQUIRED

BIL projects ERR09 and ERR10 require a DNA extractor for a five-year, research projects developing the tools of detecting invasive forest pathogens in US borders and tree nurseries. The projects involve domestic and international nurseries/gardens/arboretums that span diverse climates and woody plants. Reliable and high-quality DNA from various biological samples (e.g., fungi, plants, spores, water, soil, etc.) is essential to the success of the projects. The projects requires a one (1) DNA purification system.

Requesting organization: USDA Forest Service – Pacific Northwest Research Station

Items: KingFisher™ Duo Prime Purification System with SmartStart

2. JUSTIFICATION FOR SOLICITING FROM A SINGLE SOURCE or FOR A BRAND

NAME

Single Source Justification:

Fisher ScientificTM (ThemoFisher Scientific) is the sole manufacturer and distributor of the following equipment needed for this project:

Thermo Scientific™ KingFisher™ Duo Prime Purification System with SmartStart: Our BIL ERR09/ERR10 projects face to serious challenge for extracting DNA from forest soil samples.

We tried to use our conventional DNA extractor (tissue disruptor) for extracting DNA from these soil samples. Unfortunately, this conventional tissue disruptor resulted in no DNA or very poor yield and quality of DNA which are not usable for our purpose. It is essential to produce high quality and high yield DNA, RNA and proteins from a variety of starting samples for our BIL projects. The Duo Prime Purification System with SmartStart from ThermoFisher Scientific is providing a unique magnetic bead technology which allows the separation of DNA from crude samples such water, cultivated media, soil, fungal spores. Further, this DNA purification system can process many samples (e.g., 96 samples at a time) with automation capacity. The item being purchased will be placed at the Forest Pathology Laboratory – Corvallis Forestry Sciences Lab where the various biological samples need to be processed for the BIL projects. Due to delayed purchase of this DNA purification system, the entire BIL projects are jeopardizing to meet its deadline. High quality and high yield DNA (and later RNA and proteins) are prerequisite for proceeding our BIL projects in the next steps. We know for sure that the Duo Prime Purification System from ThermoFisher Scientific will allow us to proceed our projects to next steps – our counterpart lab (Rocky Mountain Research Station) is currently using this system to successfully process their soil (and other biological) samples. This procurement will allow PNW Research Station to meet the current BIL projects’ requirements.
