High Performance Liquid Chromatography assays developed to quantify thiamine levels in salmon muscle tissue and eggs.
The Laboratory of Dr. Jacques Rinchard, Department of Environmental Science and Ecology, SUNY Brockport Research Foundation has conducted the long-term analysis of thiamine levels in Lake
Champlain and in the Great Lakes over the past 15 years. No other lab is currently using this procedure and it would take considerable expense and time to implement this thiamine procedure in another lab and have that lab standardize with
Dr. Rinchard's lab to allow data to be added to the long-term salmon thiamine data set established in 2003.
The proposed acquisition action is for items and services for which the Government intends to solicit and negotiate with only one source under the authority of FAR 13.106-1 (b). If you believe you can provide the items or services, please forward capability statements to
Tonia_ellen@fws.gov by February 28, 2022. A determination by the Government not to compete this proposed contract acquisition based upon responses to the notice is solely within the discretion of the Government. PROJECT DESCRIPTION
Restoration efforts for landlocked Atlantic salmon (salmon) in Lake Champlain were complicated in 2003 with the illegal introduction of alewife to the lake. Alewives have become abundant in the lake and have replaced the native rainbow smelt as the primary forage fish for salmon in many areas of the lake. Unfortunately, alewives contain high levels of thiaminase, an enzyme that degrades thiamine (vitamin B1), and salmon that primary forage on alewives develop thiamine deficiency syndrome.
Salmon thiamine levels have been monitored since
2003 in the eggs of females spawned at Vermont
Fish and Wildlife Service Ed Weed Fish Culture
Station. Recent trends have indicated levels of thiamine have fallen to critically low levels that cause loss of equilibrium, weakness, and increased mortality at the swim-up fry stage.
These characteristics of thiamine deficiency syndrome have been observed over the past five years at the hatchery. Egg thiamine levels have been critically low the past five year, which requires thiamine bath treatment of eggs in the hatchery to allow fry to develop normally.
However, abundance of alewife and rainbow smelt fluctuate in the lake and there is a need to monitor thiamine levels of eggs to determine if thiamine bath treatments are necessary and assess impacts of non-native alewife on ability of landlocked salmon to reproduce naturally in Lake
Champlain tributaries.
Eggs from feral landlocked Atlantic salmon females collected at the Hatchery Brook/Ed Weed
Fish Culture Station in 2021 and 2022 will be monitored for thiamine (n = 60) n = 30 egg samples collected from feral females in Nov of 2021.
n = 30 egg samples collected from feral females in Nov of 2022.
In addition, thiamine concentrations will be monitor in egg (n = 120), muscle (n = 50) and liver (n = 50) samples from the broodstock
Atlantic salmon.
n = 20 muscle samples from White River NFH brood stocks in Nov of 2021.
