STATEMENT OF WORK
I. GENERAL INFORMATION
A. Introduction
The USGS will be performing a series of studies studying the effect of metal availability and metal limitation on cyanobacterial growth and toxicity
B. Background
Cyanotoxins are produced by cyanobacteria in freshwater ecosystems. Cyanotoxins include neurotoxins, hepatotoxins and a variety of other toxins with unknown effects. Cyanotoxins have been implicated in the deaths of wildlife, pets and livestock. In some cases, cyanotoxins are thought to be responsible for human health impacts, both acute and chronic. Not all species of cyanobacteria produce toxins, and the controls and distribution of cyanotoxins in freshwaters remains difficult to determine.
In a series of previous experiments on naturally-occurring phytoplankton communities, we observed mixed evidence of metal effects on growth and toxicity. To better understand the effects, we are looking to identify metabolic indicators of metal limitation, as well as single-species estimates of metal effects on toxin production.
To do this, we require accurate measurements of metal availability.
C. Scope:
The objective of this work is to analyze 100 samples for Fe, Mo, Ni and Zn.
II. WORK REQUIREMENTS
A. Technical Requirements
1. The USGS with perform experiments on cultures of Microcystis spp. designed to evaluate the effects of metals (Zn, Mo, Fe, and/or Ni) on growth and toxicity in a lab environment. The USGS will also collect samples to check for potential sources of contamination in the lab environment, the water supply, etc. All of these samples will be filtered and frozen and shipped frozen to the contract lab. This protocol will be discussed with the contractor, and if some other preservation method is indicated/required (e.g., acidification) the USGS will preserve samples as indicated/required.
2. The contract lab will perform SF-ICP-MS (Sector Field Inductively Coupled Plasma Mass Spectrometry) analysis of Fe, Mo, Ni, and Zn in waters. Sample analysis will be accompanied by standard quality control methods (QC), including blanks, spikes, duplicates, and reference materials (e.g., certified reference materials such as Saint-Laurent River Surface samples, SLRS). Other analytical methods that can accomplish the below detection limits will be considered.
a. There should be an expectation that the limit of detection will be at or below the values below, however we would like a contractor who is willing to determine a project-specific limit of detection and advise on materials we should use to improve our detection limits.
b. Fe – 0.355 µg/L
c. Mo – 0.0014 µg/L
d. Ni – 0.00364 µg/L
e. Zn – 0.454 µg/L
3. The analytical laboratory should function as an “Ultra-Trace Element Clean Laboratory”.
a. The clean laboratory should achieve an overall cleanliness level equivalent to Class 5000, with critical work zones operating at or below Class 10.
b. All supply air must be HEPA-filtered, and the laboratory must be maintained under positive pressure relative to adjacent spaces. An anteroom or similar buffer area should be used to isolate the clean laboratory from the surrounding building environment.
c. Air quality shall be routinely monitored, including particle concentrations and other relevant environmental variables.
d. Construction materials must be primarily non-metallic to minimize trace-metal contamination.
Any required exposed metal components must be coated (e.g., epoxy) or otherwise treated to prevent contamination.
e. Laboratory furnishings should include non-metallic cabinetry, HEPA-filtered polypropylene exhausting hoods or biosafety cabinets, additional polypropylene exhausting hoods, and high-purity water polishing systems sufficient to support ultra-trace work.
f. Sample preparation capabilities must include microwave digestion systems for complete dissolution of solid-phase materials, as well as methods for aqueous extractions, resin separations, or other low-contamination preparation techniques.
4. Provide analysis results in a digital format. Comma-separated value (CSV) files are preferred. We would like to avoid Excel spreadsheets that include any formulas. Columns should be clearly labeled and metadata explaining column labels or other aspects of the results should be provided. Results of QC work should be included in the analytical results.
5. Time standard: All results should be provided to the USGS within 3 months after samples have been received by contractors, or within a mutually agreed upon time if 3 months is insufficient.
B. Deliverables
2. All results should be provided to the USGS within 3 months after samples have been received by contractors, or within a mutually agreed upon time if 3 months is insufficient.
III. SUPPORTING INFORMATION
A. Place of Performance:
Contractor’s laboratory.
B. Period of Performance:
The period of performance shall be one year from the date of award or 90 days after the final batch of samples is received by the contractor, whichever comes first. Samples will be shipped to the contractor following experiments, sometime between July and September 2026 (exact time pending equipment availability and culture availability) and will be shipped to the contractor shortly thereafter.
C. Government Furnished Property (GFP):
None
D. Special Considerations
1. The contractor should have a trace element clean lab for analysis. This would include the ability to analyze most ultra-trace elements and metals. The staff should be experienced in working in a trace element clean laboratory environment, using standard protocols with rigorous quality assurance and quality control checks and reviews.
2. Data rights remain with the USGS. Request for use of data can be submitted to Technical POC.
