About this file

This document is a combined synopsis/solicitation for a Request for Quote (RFQ) issued by the U.S. Food and Drug Administration's (FDA) National Center for Toxicological Research (NCTR). The FDA requires protein binding and functional assay services to identify and verify drug targets associated with adverse neuropsychiatric effects. The services must analyze over 14 protein targets/receptors and provide binding, calcium flux, cAMP, and β-arrestin data in either agonist or antagonist formats. The analysis must be completed within 30 days after receipt of the test article, and the data provided in Excel and/or PDF formats. This is a one-time analysis, with no option periods. The solicitation is set aside for small business, with a NAICS code of 541714 - Research and Development in Biotechnology (except Nanobiotechnology). Statement of Work

Background The U.S. Food and Drug Administration’s (FDA) National Center for Toxicological Research (NCTR), Jefferson, AR requires protein binding and functional assay service for the targets/receptors in the human central nervous system to identify and verify drug targets that are potentially associated with adverse neuropsychiatric effects. The assays are for one compound and should be able to analyze over 14 protein targets/receptors, including 5HT1A, 5HT5A, 5HT2B, α2A-adrenoceptor, α2B-adrenoceptor, β1-adrenoreceptor, opioid μ, progesterone PR-B, dopamine D1, D3, D5, D2L receptors, and dopamine and monoamine transporters. The binding assays should be based on competitive binding with radioligands, and the functional assays should be cell-based and include calcium flux, cAMP, and β-arrestin. The assays should be performed in a dose response fashion in either agonist or antagonist or both formats.

Objectives We require protein binding and functional assay service for the targets/receptors in the human central nervous system to identify and verify targets of one drug that are potentially associated with adverse neuropsychiatric effects.

Scope

Results of radioligand binding, calcium flux, cAMP, and β-arrestin assays of the drug shall be mailto:ryan.alexander@fda.hhs.gov mailto:ryan.alexander@fda.hhs.gov delivered for 14 receptors including 5HT1A, 5HT5A, 5HT2B, α2A-adrenoceptor, α2B-adrenoceptor, β1-adrenoreceptor, opioid μ, progesterone PR-B, dopamine D1, D3, D5, D2L receptors, and dopamine and monoamine transporters. Results shall include EC50/IC50 values in either agonist or antagonist or both formats. Data provided shall in Excel and/or PDF formats, including study design, experimental conditions, and results for each target analyzed. This is test article analysis service at the contractor labs, no installation/training are needed. This is also a one-time analysis, there are on option periods.

Tasks

Analysis should be delivered within 30 days after the test article is received. All data will be provided to NCTR via email or an internet site for downloading. The service company does not own the data (i.e., intellectual property). NCTR/FDA is the owner of the data. Below are the performance requirements.

1. The assays shall be able to analyze over 14 protein targets/receptors, including 5HT1A, 5HT5A, 5HT2B, α2A-adrenoceptor, α2B-adrenoceptor, β1-adrenoreceptor, opioid μ, progesterone PR-B, dopamine D1, D3, D5, D2L receptors, and dopamine and monoamine transporters.

2. The drug targets/receptors assayed shall be human origin.

3. The binding assays shall be based on direct competitive binding with a corresponding radioligand for each target/receptor.

4. The functional assays shall be cell-based and include calcium flux, cAMP, and β-arrestin.

5. The assays shall be performed in a dose response fashion in either agonist or antagonist or both formats, and EC50/IC50 shall be calculated.

6. The assays are for one compound and shall be duplicated.

7. Data provided shall in Excel and/or PDF formats, including study design, experimental conditions, and results for each target analyzed, and all data will be provided to NCTR electronically.

Deliverables

Results of radioligand binding, calcium flux, cAMP, and β-arrestin assays of the drug shall be delivered for 14 receptors including 5HT1A, 5HT5A, 5HT2B, α2A-adrenoceptor, α2B-adrenoceptor, β1-adrenoreceptor, opioid μ, progesterone PR-B, dopamine D1, D3, D5, D2L receptors, and dopamine and monoamine transporters. Results shall include EC50/IC50 values in either agonist or antagonist or both formats. Data provided shall in Excel and/or PDF formats, including study design, experimental conditions, and results for each target analyzed.

PRICE QUOTES SHALL BE FOB DESTINATION INCLUSIVE OF ALL ANALYSIS,

SHIPPING, HANDLING, & DELIVERY CHARGES

Analysis Delivery shall occur 30 working days after receipt of order.
