TITLE

10X Genomics Single-cell and Spatial Library Prep Kit Supplies for NICHD

INTRODUCTION

THIS IS A PRE-SOLICITATION NON-COMPETITIVE (NOTICE OF INTENT) SYNOPSIS TO AWARD A CONTRACT

OR PURCHASE ORDER WITHOUT PROVIDING FOR FULL OR OPEN COMPETITION.

The National Institutes of Health, National Institute of Child Health and Development (NICHD) intends to award a purchase order without providing for full and open competition to GOVERNMENT SCIENTIFIC SOURCE INC for 10X Genomics Single-cell and Spatial Library Prep Kit Supplies.

BACKGROUND

Transcriptome analysis at single cell resolution (single cell RNA-seq) has led to significant advances in our understanding of biology, elucidating cellular heterogeneity in complex systems and tissues, and enabling discovery of novel cell types, states, and biomarkers with relevance to clinical applications. 10x Genomics has developed Chromium Single Cell (sc) Multiome ATAC + Gene Expression, a fully supported turnkey solution with assay and software components. This product provides researchers with a unified view of a cell’s open chromatin landscape and gene expression profile, it is possible to resolve how cell types and states are established, discover new gene regulatory interactions, and interpret epigenetic profiles with key expression markers.

Efforts to link epigenetic and transcriptomic measurements from single cells have historically necessitated two separate assays. This requires two different input samples followed by inferred linkage between separate gene expressions and chromatin accessibility (ATAC-seq) datasets. In addition to the computational challenges of accurately inferring and linking similar cells across samples, this approach may be affected by different cellular proportions between input samples and different sample preparation conditions that can influence data output and quality. This is the only commercially established solution for simultaneous capture of both transcriptomic and epigenomic modalities in the same single cells, across thousands of cells.

This technique avoids the computational errors associated with linking separate datasets and maximizes possible readouts from precious samples. These products are relied upon for the work of dozens of NICHD DIR investigators, including Sheppard lab. Without these purchases the NICHD-DIR projects will be stalled and severely impacted. There is no alternative product available in the market that will be used on the sequencers.

JUSTIFICATION

This is a Sole Source request because 10X Genomics is the only company that can provide the reagents and maintenance contracts that are compatible with our 10X Genomics CHROMIUMX controller, VISIUM and XENIUM instruments. The Chromium, Visium and Xenium instruments are sophisticated machines that is used by many NICHD investigators and needs the specific reagents requested. The 10 X Genomics technology offers sequencing-based and imaging-based solutions for single-cell transcriptomics and multiomics. MGC provides service for all NICHD researchers to unlock a full spectrum of genetic variation and biological function with these cuttingedge technologies. The reagents, instruments and service contract requested are provided only from this company. Reagents from other companies are not compatible.

REGULATORY AUTHORITY

The resultant contract will include all applicable provisions and clauses in effect through the Federal Acquisition Regulation (FAR) Circular (FAC) 2025-05 effective 08/07/2025. This acquisition is conducted under the procedures as prescribed in FAR subpart 13-Simplified Acquisition Procedures at an amount not exceeding the simplified acquisition threshold ($250,000) and FAR Subpart 12- Acquisition of Commercial Items.

This acquisition is conducted under the authority of FAR 13.106(b)(1) Soliciting from a single source - For purchases not exceeding the simplified acquisition threshold.

DESCRIPTION OF REQUIREMENT

We require sc-Multiome library kits and flowcells for sequencing samples in a timely manner and compatible with our Illumina Novaseq 6000 and MiSeq sequencers. This purchase is essential to maintain current research activities with minimal disruptions.

CLOSING STATEMENT

This synopsis is not a request for competitive proposals. However, interested parties may identify their interest and capability in response to this notice and may submit a capability statement, which shall be considered by the agency. Responses to this notice shall contain sufficient information to establish the interested parties’ bona-fide capabilities for fulfilling the requirement. A determination by the Government not to compete this proposed contract based upon responses to this notice is solely within the discretion of the Government. The information received will normally be considered solely for the purposes of determining whether to proceed on a non-competitive basis or to conduct a competitive procurement.

All respondents must have an active registration in the System for Award Management (SAM) www.sam.gov.
