About this file

This document is a federal solicitation (RFQ-NIAID-24-2223252) for a brand name or equal digital surface plasmon resonance (SPR) equipment called the Alto16 Channel Instrument with Nicosystem Pro Software. The National Institute of Allergy and Infectious Diseases (NIAID) has a requirement for the Alto16 instrument, along with associated cartridges, reagents, training, and warranty/service. The solicitation is a full and open competition, with no small business set-aside. Quotes are due by May 24, 2024 at 4:00 PM EST, and the award will be based on technical capability and price. The solicitation specifies the required Alto16 instrument and accessories, and provides details on the instrument's capabilities, such as digital microfluidics, 16 optical channels, and ability to measure binding kinetics. The NAICS code is 334516 (Analytical Laboratory Instrument Manufacturing) with a size standard of 1,000 employees.

Alto16 and components SPEC SHEET Brand Name or Equal to these minimum requirements:

System contains all of the fluid management in a disposable, single-use cartridge that uses digital microfluidics and integrated plasmonic SPR sensors.

· Alto uses a combination of Nicoya Lifesciences Inc.’s unique digital microfluidics platform and surface plasmon resonance technology to measure binding kinetics between biomolecules.

· Alto™ utilizes digital microfluidics, a disposable fluidics technology that eliminates the need for fluidics cleaning cycles and risks associated with clogging.

· Alto features 16 independently addressable optical channels and a built-in computer to operate the digital microfluidics powered instrument and acquire binding data.

· Alto cartridges are an integrated consumable that are powered by digital microfluidics, and feature both sample/reagent wells and 16 fiber optic sensors with gold nanostructured surfaces.

· Binding kinetics are acquired by immobilizing ligand biomolecules on the cartridge's 16 sensor surfaces. Label-free analyte biomolecules in solution interact with the immobilized biomolecules, and the degree of molecular binding is quantitatively measured through the instrument's optical detection system.

· Alto cartridges provide a complete suite of experimental automation features including: automated serial dilutions, highly accurate liquid droplet dispensing, and automated mixing of samples.

· Alto is compatible with proteins, DNA, carbohydrates, lipids, certain small molecule applications, antibodies, virus-like particles, nanoparticles, serum, lysate and more sample types. Due to the digital microfluidics technology, Alto requires only 2µL of sample and eliminates the risk of sample evaporation during experiments.

Alto Specifications

· Alto utilizes 2µL of sample and can perform five 3-fold serial dilutions of samples automatically.

· Experiments run on Alto can be temperature controlled at 15oC, 25oC and 37oC.

· Kinetic association rates up to 1 e9 1/(M*s) can be measured with Alto.

· Kinetic dissociation rates between 1e-5 to 1.0 (1/s) can be measured with Alto.

· Equilibrium dissociation constants between a range of 1e-12 to 1e-3 (M) can be measured with Alto.

· Alto kinetics cartridge can contain up to 16 unique surfaces for sample ligands and 48 sample analytes on a single kinetics cartridge.

· Alto has dimensions of 1.5H x 1W x 1.5D ft (46H x 35W x 51D cm) and weighs 51 lbs (23 kg).

· Alto has a near instantaneous transition time (<0.1s) between buffer and sample.

Control and Analysis Software

· Alto is controlled by cloud or local based software that guides the user through all operational steps including sample loading, experimental setup, optimization, and data analysis.

· Raw binding data is recorded in real-time on Alto software and can be analyzed using an external computer via the local or cloud-based software.

· Binding kinetics, quantitation, screening and epitope binning applications provide the user with data analysis, curve fitting, and quantitation algorithms.
