About this file

This document summarizes a request for quotes (RFQ) from the National Institute of Allergy and Infectious Diseases (NIAID) for peptide synthesis services spanning the entire tuberculosis proteome. The NIAID Vaccine Research Center seeks a firm-fixed price contract to create, validate, and pool approximately 132,000 peptides over an 18-month period of performance. Quotes are due by September 2, 2021, with award anticipated on September 10, 2021. This opportunity is set aside for small businesses. The selected vendor must complete at least 50% of the work, with up to 50% allowed to be subcontracted. Quotes should include separate volumes for pricing and technical approach, and address the vendor's capability to meet the statement of work requirements. The award will be made to the responsible offeror representing the best value based on technical approach, past performance references, and price.

QuestionGovernment Response
1We would prefer to bill on a monthly basis, for work completed. Is this agreeable? If not, what terms for billing are preferred? This work is for a single outcome that cannot be subdivided into separate, severable performance deliverables. For this reason, it is not possible for there to be monthly invoices based on complete work.
230% purity is referenced as a requirement. It is also referenced that mass spec would be the form of data preferred. Mass spec isn’t as quantitative as HPLC and a mass spec would be. We have both capabilities, but we want to ensure we are quoting what is asked for. Would the combo of the two be preferred, or is only mass spec needed?Whichever method the vendor feels provides a good estimate of purity is fine.
3The pools are stated to be between 1 and 70 peptides. 70 peptides means that the pool would be 350 mg (70 x 5 mg). Each pool will be aliquoted into 5-20 vials/plates. This may pose a volume problem in this format. We would likely need to investigate a higher volume 96 well plate vs our standard 1.3 ml vials. Is there a higher volume 96 well plate in mind for this work? Or is this something that you would like us to pursue options for? As an example, if we take the 350 mg/20 it is 17.5 mg of peptide in each vial per plate. This may be difficult to keep in solution at the very high concentration needed to aliquot and fit into the small 96 well plate vials. Any additional information on plates and aliquoting would be beneficial for us to quote accurately.A large volume format for 96 wells (e.g. 2mL deep well) would be fine. We can also have more master plates prepared if that helps.
4What information will be needed on the individual vial labels? Or will all information be contained on spreadsheet only? The individual vials should be labeled with the pool ID.
5Given the fact the number of peptides to synthesize for full coverage of the proteome is huge, it is almost a certain that some peptides will fail synthesis or QC especially the hydrophobic ones. In these cases, is it acceptable to skip those? Please clarify. If repeated attempts at synthesis fails, and the vendor is unable to suggest an appropriate alteration of the peptide ends (e.g. moving the break point to alleviate hydrophobicity) then skipping the peptide is acceptable.
6Will we be assigned all the sequences one time or is the plan to provide sequences in batches? Or is it for us to figure out the sequences and deliver within 18 months on set schedule? All proposed sequences and pooling information will be supplied in advance. Vendor may suggest alternative breakpoints as per their expertise.
7Please clarify the policies or any specific requirements for any subcontracting of difficult to synthesize peptides to our sub-contractors both (local and overseas) to take advantage of their specialized experience for difficult to synthesize peptides towards successful completion of the project. There is a limitation on how much of the work can be subcontracted. Since this is a small business set aside, you must complete at least 50% of the work. Make clear in your quote any potential subcontracting (what vendor and for what work, including the percentage of the contract value that would be subcontracted).
