Part 3 – Description/Specifications

3.0 Background

The requested system is needed to prepare and analyze samples using Site-specific Nuclear Isotope Fractionation (SNIF) by Nuclear Magnetic Resonance (NMR). The system shall have the capability to prepare food samples containing sugar and/or ethanol for analysis to determine stable isotope ratios for deuterium/hydrogen and carbon-13/carbon-12. These are the most commonly used nuclei for SNIF-NMR. The system shall be able to obtain a consistent and repeatable result and therefore shall feature the modules listed below. The method involves measuring the isotopic ratios of the hydrogen and/or carbon of the ethanol that is either present in the sample or analytically derived therefrom. The ratios are then used to compare authentic and/or standard materials to samples of unknown or uncertain provenance. As this measurement is very sensitive to the exact ratios, the process shall be very well controlled and coordinated between all the steps of sample preparation and analysis. These measurements are valuable in many instances as they offer information about the source of a material that may not be available by any other means. This process requires several steps that shall be handled by the various modules of the SNIF system listed.

Specific Requirements:

1. The system shall have the ability to completely convert sugars in the sample into ethanol by fermentation. A module is needed for monitoring fermentation to ensure that 100% of sugars are converted to ethanol during the fermentation. This shall consist of a refractometer, an analytical balance, and a dedicated PC with software for monitoring and reporting to a database. It shall ensure that there is no isotopic fractionation during the fermentation of sugars from the samples.

2. The system shall have the capability of distilling the samples to 95% ethanol. This requires a distillation unit equipped with Cadiot columns with temperature control, a balance and any components required to ensure maximum distillation. It shall monitor and integrate with the other components to ensure no isotopic fractionation from the distillation step. The ADCS Windows, the distillation module listed by Eurofins, are offered in either a 4- or 8-window option. The 4-window option is all that is required; this 4-window option shall be included in the module.

3. The system shall be capable of comparing the alcohol content before and after fermentation. This shall be accomplished with an electronic densimeter with a controlling computer and software to interface with a database to compare data with the other modules.

4. The water content shall be accurately determined with a Karl Fischer unit and a dedicated analytical balance with a controlling computer and software. This shall be done in conjunction with the density measurement.

5. The system shall have a computer and software to interface with an NMR spectrometer to transform the signal and calculate the integration of the peaks in the spectrum for each sample. The system shall interface with the other modules to calculate the isotope ratio for each site based on the precise measurement of the ethanol content and the spectroscopic data.

6. Finally, a computer with software to integrate all the modules and data shall be used to ensure accuracy and precision throughout the process and throughout the service life of the system.

All of this shall require detailed training, installation and validation to ensure system is functional and up to the specifications needed to perform official methods based on the SNIF-NMR technique. All the components listed above shall be required to properly perform the method and they need to be integrated to ensure the quality of the data.

3.3 Reporting Requirements and Deliverables:
