Part 3 – Description/Specifications
3.0 Statement of Work:
The FDA is looking to purchase an automated protein stability analyzer that is capable of quantifying and predicting protein stability across a wide range of biochemical conditions.
3.1 Background
The FDA is looking to purchase an automated protein stability analyzer that is capable of quantifying and predicting protein stability across a wide range of biochemical conditions. Our current need is in support of bioprocess manufacturing experiments in which a new model IgG1 is produced and subsequently purified and characterized using gold standard protein analytical equipment. In order to keep up with current manufacturing capabilities, the FDA requires that we stay current in both technology and techniques. The instrument being requested has the unique ability to test a single protein across a wide range of conditions while monitoring denatured, aggregated, and denatured/aggregated composition in any particular biochemical formulation. This allows protein stability to be both quantified and predicted in future conditions for the first time in a fully automated assay. Data collected from this instrument in 1 day would require countless man hours, extended incubation times, and numerous other analytical instruments. A small footprint and simple loading procedure ensure that human error is minimized and the most accurate protein stability data is collected.
3.2 Technical Specifications
1. Instrument shall be able to fit on the bench top with minimal foot print (<60cmW x 75cmD x 88cmH) to allow for its use to be efficiently incorporated in to the current laboratory space
2. Instrument shall include: automation and fluidics module, fluorescence detector, workstation, software, and 1 tray (blue).
3. A 12 month warranty shall be included in the initial purchase which would include replacement of damaged parts as well as technical support.
4. Onsite training shall include experimental set up, system operation and basic data analysis.
5. Startup consumables shall be included in the initial purchase to include vials, tops, and plates appropriate for the selected included tray.
6. Shall be able to hold protein samples at 4°C throughout assay.
7. Shall be able to fully automate the mixing and measuring of protein aggregation/denaturation in a high-throughput (96-well), minimal time (~1 hour/condition) format.
8. Shall be able to perform all assays using minimal protein content ( <50 µg/mL).
9. All items shall be deliverable in 6 months or less
3.3 Items required
1. Protein Stability Analyzer base unit (equivalent to HUNK p/n 300-1029) with 1 operating tray (similar to HUNK P/N 302-1034 Blue Tray)
2. Installation and training
3. One (1) additional day of onsite application training
4. Shipping costs
5. Startup assay consumables including:
a. 96-well microplates (similar to HUNK P/N 301-1044)
b. Deep well microplates (similar to HUNK P/N 301-1045)
c. Compatible protein vials with caps (similar to HUNK P/N 301-1050)
