3.0 Background

The FDA requires a Fluorescence Protein Thermal Stability Measuring Instrument (Unit).

Using the instrument we would like to measure in a non-condensing relative humidity range of 40-60%:

· Intrinsic fluorescence of proteins in a very small volume (9 µL per sample)

· Fluorescence of polypeptides or proteins in a concentration range of 100 µg/mL to 150 mg/mL

· Fluorescence of some extrinsic dyes incorporated to proteins

· Temperature scans of fluorescence at a rate of 0.01 °C/min to 10 °C/min

· Melting temperature of polypeptides and proteins

· Aggregation temperature of polypeptides and proteins

· Thermal stability analysis

· Multi-sample analysis, as many as at least 48 samples in one hour.

Protein aggregation is an important trigger of immunogenicity in therapeutic proteins treatments. Thermal stability and unfolding studies play an important role in determining the propensity of the proteins to aggregate in specific formulations. DPQR will use the UNit to understand the model therapeutic proteins thermal stability in various buffer conditions as a way to determine optimum composition for improving the quality of therapeutic or model proteins. Therefore, DPQR requires one (1) UNit instrument with specifications outlined in the ‘Technical Evaluation Criteria’.

DPQR requires one (1) instrument with the following specifications:

An apparatus to Measure Thermal Stability Using Intrinsic Fluorescence of Proteins

· A bench top compact instrument for protein thermal stability determinations in a user friendly, easy, automated manner.

· The instrument determines the melting points of proteins in a micro volume and high throughput analysis set up using intrinsic fluorescence.

· Is capable of simultaneously measuring fluorescence and light scatter of the samples.

· The software is capable of measuring the Tm (melting temperature) and onset of Tagg (aggregation temperature) of proteins.

· Data should be exportable in a number of formats as may be required.
