PART 2 – DESCRIPTION OF REQUIREMENT
BACKGROUND INFORMATION
In preparation for the Multi-Lab environmental CORE samples and method implementation of DFPG# 20-14
Romaine Lettuce Sampling Assignment for rt PCR detection of Diarrheagenic Escherichia coli 0157 or
EHEC/STEC analysis in food and environmental samples. Identical instrumentation is needed in each participating field laboratory that is capable and optimized for performing this technique. Use of this method for EHEC/STEC analysis is needed due to its increased sensitivity and accuracy over traditional manual techniques.
Offeror Name DUNS http://inside.fda.gov:9003/downloads/ProgramsInitiatives/Food/FieldPrograms/UCM643717.pdf http://inside.fda.gov:9003/downloads/ProgramsInitiatives/Food/FieldPrograms/UCM643717.pdf
OBJECTIVES
The objective for this work request is to provide instrumentation that is capable and optimized for the implementation of the analytical bead retrieval method described in FDA Bacterial Analytical Manual, Chapter
4A Diarrheagenic Escherichia coli for the following participating field laboratories: ARK, DEN, NFF, PSF, SAN & SFF.
SCOPE
The Contractor shall provide UPLC instrumentation with identical technical specifications and functional features as the Acquity Classic UPLC on which the method was developed and validated. Services shall include delivery of the instrumentation to the laboratories as specified, as well as on-site installation, qualification, training, and standard equipment warranties.
The Contractor shall provide six (6) new KingFisher (or equivalent) Food Protection Purification System
Contractor shall provide delivery of the instrument to the FDA Laboratories as listed below and provide on-site installation and performance qualification by a qualified service technician.
Contractor shall demonstrate upon installation that all items meet performance specifications and provide documentation of required qualifications (IQ/OQ/PQ) at completion of installation. Instrument shall not be accepted until the specifications have been met and the qualifications performed.
Contractor shall provide on-site instrument familiarization to the analysts who will be operating the instrument.
Contractor shall provide a warranty for each entire system for parts and labor for 12 months from the date of installation acceptance.
BRAND NAME OR EQUAL SALIENT CHARACTERISTICS
FDA Bacterial Analytical Manual, Chapter 4A Diarrheagenic Escherichia coli describes a rtPCR based method of rapidly detecting EHEC/STEC in food and products using a magnetic beads to isolate genes specific to
EHEC/STEC. This method vastly increases the sensitivity and throughput as compared to the manual methods currently in use. This method has been validated at the Single Laboratory level and is now scheduled for implementation at all FDA Food and Feed analysis laboratories.
The method has been validated and demonstrated to be optimally performed on the Food Protection Purification
System which has unique features that are integral to method performance:
The custom-built magnetic particle processor is designed for the automated transfer and processing of magnetic particles in a 1mL tube well scale.
Functions without any dispensing or aspiration parts or devices.
The automation of manual steps enables simultaneous processing and purification increasing the sample throughput.
Enables a quicker reaction and a more efficient wash due to the simultaneous shaking during incubation and washing.
Facilitates a good collection of bead-bound sample due to the efficiency of the magnet and prevents carry-over due to the fact that the same tip is used for one sample.
Facilitates the whole processing with the aid of an internal program.

The entire system(s) must be warranted for parts and labor for 12 months from the date of installation. The vendor must also be capable of servicing the instrument through the extended warranty or service contract. The system must include at least a one (1) year warranty and shall include at a minimum: coverage on all non-consumable items and parts supplied including base instrument, computer, monitor, factory-certified replacement parts, engineer labor, and travel costs. This requirement does not obligate the Government to purchase an extended warranty.
FOB point destination to include inside delivery, set-up, installation, clean-up of area, and familiarization training. Sufficient training for up to four users will be provided during installation, such that operators may independently operate the instrument and increase familiarity and proficiency. The training shall include manuals and consumables to be used during training.
The contractor provides for installation qualification (IQ/OQ/PQ) and provide associated documentation at completion of installation.
DELIVERY OF SERVICES
The delivery of the system and installation/qualification/training are requested to be completed by: 30 days after award
