About this file

This document is a User Requirement Specification (URS) issued by the Dakota Bioproducts Innovation Institute (Dakota BioWorx) and South Dakota State University, with funding from the U.S. Department of Commerce Economic Development Administration (EDA). The project involves the procurement, delivery, and installation of five primary research sterile bioreactor systems for process screening and workforce development. Each unit will consist of four 1L or less bioreactors with independent controls, operated from a single control unit. The systems must meet various technical requirements related to construction, health/safety, operation, materials, and reliability. Bids should be returned to the specified contacts by the due date.

The selection criteria include mandatory technical requirements, pricing, technical response, lead time, warranty, and vendor experience. Vendors must comply with federal "Buy America" and anti-boycott provisions. The contract will be partially funded by the EDA, and the successful vendor must provide system validation documentation, startup assistance, and access to US-based service and spare parts.

2 SCOPE

The project will include a primary research sterile bioreactor system for screening process conditions, in addition to assisting in workforce development. This document defines the requirements and deliverables for five of these systems. It describes overall requirements that must be met to produce the specific equipment requested.

This document covers the procurement, delivery, and install of the systems.

This URS is the input document for the:

• Equipment procurement purposes

• Equipment sizing

• Functional and technical specifications

3 BACKGROUND

Dakota Bioworx is a research facility for the development of high-quality bioproducts.

Primary research bioreactors will be installed to enable customers to scale up their processes and assist in workforce development. These bioreactors will not be used as seed vessels; all use will focus on screening operating conditions at small scale and training workforce on proper bioreactor practices.

4 PROCESS DESCRIPTION

A single unit, consisting of four bioreactors, will be installed. Each bioreactor will have independent agitation, pH, temperature, and aeration controls but be operated from a single control unit. Bioreactors will be appropriately sterilized via autoclave. These systems will be commercial grade that are easily configurable to handle a wide variety of process requirements. This document specifically covers systems with reactor volumes of 1L or less.

5 BASIS OF DESIGN

5.1 Capacities

One unit shall consist of four bioreactors with a working volume of approximately 1L or less.

5.2 System Construction

The unit will have four vessels monitored by a single control tower.

5.3 Health, Safety and Environment (HSE)

The unit will satisfy appropriate conformity assessment procedures and the controls shall carry the UL marking. Systems shall be constructed to adhere to proper aseptic technique.

5.4 Operation, personnel and automation

The unit will be fully automated such that operations personnel will not need to attend to them once they are started and operating.

5.5 Materials of Construction

All materials of construction must be resilient to typical chemical cleaning agents such as phosphoric acid, potassium hydroxide, and sodium hydroxide up to 3%.

5.6 Reliability & maintenance

The fermentation systems will be designed to operate routinely during 24 / 7 / 365 operations. All wear parts shall be easily accessible and replaced on a regular maintenance schedule. Wear parts like gaskets shall be stock items that can be ordered and delivered in a timely manner.
