Notice of Funding Opportunity Purpose
The purpose of this notice of funding opportunity (NOFO) is to support a consortium of Translational Centers using Microphysiological Systems (MPS) for Infectious Diseases (TCMID). Recent efforts in the MPS fields are moving towards validation and regulatory use but have not focused upon models studying infectious diseases and testing of medical interventions. The NIH and FDA have recently issued guidance and established initiatives for enhancing use of New Approach Methodologies (NAMs) in research and product development. This includes in vitro systems that allow scientists to model human disease and capture human variability and patient-specific characteristics that will accelerate safer treatments for patients while reducing costs and sparing use of laboratory animals. To meet this need, the Centers will refine well validated human tissue models studying infectious diseases to a level consistent with regulatory qualifications that will promote their adoption for use by product developers, supporting regulatory acceptance.
Funding Opportunity Goal(s)
To assist public and private nonprofit institutions and individuals to establish, expand and improve biomedical research and research training in infectious diseases and related areas; to conduct developmental research, to produce and test research materials. To assist public, private and commercial institutions to conduct developmental research, to produce and test research materials, to provide research services as required by the agency for programs in infectious diseases, and controlling disease caused by infectious or parasitic agents, allergic and immunologic diseases and related areas. Projects range from studies of microbial physiology and antigenic structure to collaborative trials of experimental drugs and vaccines, mechanisms of resistance to antibiotics as well as research dealing with epidemiological observations in hospitalized patients or community populations and progress in allergic and immunologic diseases. Because of this dual focus, the program encompasses both basic research and clinical research.

Section I. Notice of Funding Opportunity Description
Research Objectives This notice of funding opportunity (NOFO) will establish a consortium of Translational Centers using Microphysiological Systems (MPS) for Infectious Diseases (TCMID) that will have the following goals:
Refine human cell-based MPS models of infectious diseases that are fit-for-purpose for advancing study of medical interventions
Establish specific endpoints in the MPS models, focused on regulatory validation requirements, that define human pathophysiological responses to infection as indicators of therapeutic efficacy (e.g., endpoints, biomarkers, etc.)
Establish 2-4 specific Contexts of Use (COU) for each Center, with the goal of submitting to the United States (U.S.) Food and Drug Administration (FDA) a letter of intent (LOI) targeting the Drug Development Tools (DDT) Qualification program or the Fit-for-Purpose Initiative.
Structure of the Translational Centers Using Microphysiological Systems for Infectious Diseases Program Each Center in the TCMID consortium will be organized around an investigator-selected theme focused upon use of MPS as preclinical models of infectious diseases for product development. Examples of Center themes could include, but are not limited to, the following areas:
Projects focused on developing individual MPS platforms focused on infection models with different pathogens.
Projects focused on developing different tissue MPS platforms that model infections for a particular species/strain or related pathogen.
Administrative Core The Administrative Core is responsible for the management, coordination and supervision of all Center and consortium activities. A Scientific Advisory Board (SAB) will serve as an independent, external advisory body for the TCMID, but will not be involved in the day-to-day activities of the Center. All recipients will form a TCMID consortium, which will meet annually. The Core will also coordinate annual individual Center site visits from NIAID staff. Further, recipients will also become members, and participate in, annual meetings of the NIH Tissue Chip Consortium.
Qualification Core The Qualification Core is responsible for the management, coordination and supervision of regulatory-related activities. Responsibilities include coordination of activities between the Center and regulatory officials to refine COUs as needed, and leading discussions of project progress and COU to meet Qualification or Fit-for-Purpose initiatives requirements, such as with FDA's Innovative Science and Technology Approaches for New Drugs (iSTAND) program.
Scientific Core(s) The MPS Scientific Core(s) are responsible for ensuring that standards, authentication and best practices are followed for resources, such as the use of robust and reliable cell sources, well-characterized challenge material, validated MPS hardware, and instrumentation. The MPS Scientific Core(s) may also establish collaborations with other academic researchers, clinicians and industry as needed for the work. Scientific Core(s) are intended to only serve the needs of Center Research Project investigators; they will not conduct research independent of the served Research Projects.
Research Projects Research Projects will advance MPS infectious disease models and target one or more pathogens. Each Research Project will be directly linked to at least one specific COU and contribute to the Center theme and objective(s).
Applications Not Responsive to this NOFO The following types of applications are not responsive to this NOFO and will not be reviewed:
Projects that do not propose human-derived MPS platforms to directly study human-relevant infectious diseases
Projects that propose to use MPS platforms with no demonstrated robust technical reproducibility
Projects that propose animal studies
Projects that propose to use MPS platforms without fluid flow
Projects that propose the use of MPS platforms without at least two COUs (one for each Research Project)
Projects developing MPS platforms that use animal-derived cells or that do not use primary human cells or human derived induced pluripotent stem cells (iPSCs)
Projects developing (or focused on) radiological or toxin testing platforms
Projects that do not utilize a virulent form of pathogen(s)
Models without direct infection with pathogen(s), with the exception of those utilizing lymphoid tissue models with, at minimum, functional antigen-presenting cells (APC) and T and B lymphocytes for demonstrating quantitative antigen-specific immune responses to vaccine exposure
Note: Applicants are strongly encouraged to utilize pathogens that are representative of clinical isolates.
