Brand Name Justification for Other than Full and Open Competition at or Below the Simplified Acquisition Threshold
Project Title: Acquisition of 10x Genomics Library Construction Kits I. Identification of the agency and contracting activity
| DHHS, NIH, NIAID, LISB | |
| Address of the requesting lab/office: | 4 Memorial Dr. |
Building 4 Room 135 Bethesda, MD 20892
A. Project Background:
The LISB is studying the response of the immune system to the SARS-CoV-2 virus on a global scale, and at single cell resolution. The 10x Genomics single cell platform allows chromatin profiling of tens of thousands of single cells in parallel to see how chromatin compaction and DNA-binding proteins regulate gene expression at high resolution in an easy to use platform, consisting of the 10x Chromium Controller hardware, and the companion kits of reagents and consumables that allow use of the system. To help understand the host immune response to SARS-CoV-2 infection, a consortium of labs at the NIH are performing a systems-level characterization of various cohorts of patients with severe COVID-19.
Project Description:
These reagents will be used on an existing NIH-owned 10x Chromium Controller to examine the transcriptome state of millions of blood immune cells from COVID-19 patients. These data will allow the dissection of cell types, cell states, and gene-gene networks that underly immune responses in health and disease.
Description of the supplies/services to be purchased: Animal change stations will be used in bldgs. 13 and 32 RML RMVB A. Name of manufacturer, producer, or distributor of each brand name product referenced (and address if not well known):g Manufacturer: 10X GENOMICS Pleasamton, CA
Identification of the statutory authority permitting other than full and open competition (SELECT ONE)
☒ 41 USC 3304(a)(1) and FAR Part 6.302-1: Only one responsible source / Brand Name
II. Demonstration that the proposed contractor(s) unique qualifications or the nature of the acquisition requires use of the authority cited.
A. Name and address of the proposed contractor(s):
10X Genomics, Inc.
7068 Koll Center Parkway Suite 401 Pleasanton, California 94566 United States
Demonstration that the proposed brand name product’s salient characteristics (specific features, qualities and/or the nature of the acquisition) requires use of the authority cited:
10X Genomics offers proprietary microfluidic circuits that rapidly partition input samples (e.g. higher molecular weight DNA, single cells), which are then barcoded using the GemCode Technology. This technology is not available through any other vendor or instrumentation. This is the only known instrument in existence that meets our need to sequence thousands of transcriptomes is a single run from single cells and while obtaining phenotypic data from those same cells.
No other kits/reagents are compatible with instrument and attempting to run another vendor’s similar product would not be supported by the instrument. This could cause the instrument to become non-functioning and un-repairable. Therefore, having to replace equipment would be very costly to the government. Members of the DCIA consortium have used to 10X GemCode Technology and the current request for 10X Chromium Library Construction kits and 10X Chromium Controller instrument will ensure on-going research/experiments are not compromised with changing variables. Maintaining existing reagents/protocols is paramount to the success of on-going research efforts and the goal of the DCIA-funded initiative to provide a standardized toolkit to the NIH research community for investigating antigen-specific B-cell responses.
