About this file

This document is a Purchase Description for a federal contract opportunity to provide Custom Antibody-conjugated drugs (ADC) services. The objective is to generate two custom ADCs (antibody vc-MMAE conjugates) within 10 weeks of initiation, using antibodies supplied by the vendor. The minimum quantity required per ADC is 50 mg, delivered in a physiological buffer at a defined concentration with low aggregation.

The related federal contract opportunity is Solicitation Number 75N95024Q00376, issued by the Department of Health and Human Services' National Institutes of Health, National Institute on Drug Abuse. This is a small business set-aside requirement for Custom Antibody-conjugated drugs (ADC) services. The Government anticipates awarding a firm-fixed price purchase order with a 30-day delivery after receipt of order. Responses are due by July 29, 2024 at 12:00 pm EST and must include evidence of technical capability, past performance, price, and compliance with administrative requirements.

Version 11-30-2016-1 FAR Part 11—Describing Agency Needs

PURCHASE DESCRIPTION

1. Statement of Need and Purpose: Custom Antibody-conjugated drugs (ADC) services

2. Background Information and Objective:

Antibody-conjugated drugs (ADCs) are a targeted therapeutic strategy used to treat diseases of aging by combining the specificity of antibodies with the potency of cytotoxic drugs. The antibodies in ADCs are designed to recognize and bind to specific antigens that are overexpressed on the surface of aging or diseased cells, such as in certain cancers or senescent cells. Once bound, the ADC is internalized by the cell, where the cytotoxic drug is released, selectively killing the diseased cells while minimizing damage to healthy cells. This targeted approach enhances treatment efficacy and reduces side effects, addressing age-related diseases with greater precision and safety.

The application of custom antibody-directed cytotoxicity holds immense promise in addressing the need to target and eliminate senescent cells in vivo within the realm of vascular senescence research. Senescent cells accumulate in aging tissues and contribute to age-related vascular pathologies, necessitating innovative therapeutic strategies for their removal. Custom antibodies engineered to specifically target surface markers or antigens expressed by senescent cells can be conjugated to cytotoxic effector molecules, such as toxins or immune cell-recruiting agents. This approach enables precise recognition and destruction of senescent cells while sparing healthy cells, thus mitigating potential off-target effects. By harnessing the power of antibody-directed cytotoxicity, researchers can develop targeted therapies to selectively eradicate senescent cells in vascular tissues, offering promising avenues for combating age-related vascular diseases and promoting healthy aging.
