Justification for single source acquisition (what is necessary and unique about the product, service or source; steps taken to confirm unavailability of competition, as appropriate):
The FlowCam® is a unique digital imaging particle analyzer that allows the user to image, enumerate and classify particles in a continuous flow medium. One of its primary uses is the quantification of marine and freshwater plankton for academic research and water quality monitoring.
FlowCam technology is covered under various U.S. and international patents, and Yokogawa Fluid Imaging Technologies, Inc. is the sole manufacturer and sole distributor of the FlowCam in the United States and Canada. There are no alternative authorized dealers in these countries. More information about all our products and applications can be found at www.fluidimaging.com. Our product line includes the FlowCam 8000 series (FlowCam 8100 and 8400), FlowCam 5000, FlowCam Macro, FlowCam Nano, FlowCam LO, and FlowCam ALH.
Unique attributes of the FlowCam 8100 and 8400 (including the FlowCam Cyano) include:
• Ability to image particles in a continuous flow, meaning that it does not require sheath fluid, allowing the user to image multiple particles in a single frame using Autoimage mode.
• Ability to configure the instrument to acquire color or monochrome (i.e. black & white) images with a camera resolution of 1920x1200 pixels.
• Ability to image and enumerate particles ranging in size from 2 μm to 1mm using up to 4 interchangeable magnifications and corresponding flow cells.
• Sample processing speed ranging from 0.05 mL/minute at the highest magnification (20X), and up to 5 mL/minute at the lowest magnification (2X).
• Autofocus feature that allows the user to focus the FlowCam automatically prior to initiating a sample run.
• VisualSpreadsheet®, the FlowCam’s proprietary software, includes computer algorithms that acquire particle images, morphological information, and fluorescence data for each particle imaged. VisualSpreadsheet offers the capability of semi-automated particle identification and classification based on an image recognition algorithm that analyzes the 40+ image parameters collected on all particle images.
Unique futures specific to the FlowCam 8400 (including the FlowCam Cyano) include:
PUR 7776 (Revised 07/2022) Page 4 of 9 Incorporated by Reference: Rule 60A-1.045, F.A.C.
• Choice of one of 3 laser wavelengths (532 nm, 488 nm, or 633 nm) and two corresponding channels of fluorescence detection, including one for chlorophyll.
• A camera triggering mechanism whereby particle fluorescence triggers the camera to record a particle image. Fluorescence-based image acquisition allows the user to isolate images of organisms containing chlorophyll and other pigments from other non-fluorescing particles that may be of no interest to the user. The FlowCam 8400 is unique in its ability to allow the user to choose to process samples in Trigger Mode (with fluorescence) or Autoimage Mode depending on what is most appropriate for the sample.
The FlowCam 8400 is the only device to provide fluorescence data along with visual imaging (color or monochrome) of particles in a continuous flow.
Sole Source Justification and Specifications for the Acquisition of a Flow Cytometry Instrument The Florida Department of Environmental Protection, Biology Program, seeks to augment its taxonomic identification capabilities with the acquisition of an efficient flow cytometer for freshwater cyanobacteria and algal identification. The flow cytometer instrument is needed to handle the ongoing shortfall of analytical capacity created by a consistently increase of bloom response workload, cooperative bloom monitoring agreements, competing agency priority projects, and expertise gaps due to staffing turnover. To adequately meet the needs of the biology laboratory, it is imperative to obtain instrumentation to provide enhanced analytical capabilities.
The flow cytometry specifications, computer software, and data file format for the new instrument must be compatible with our existing methods to maintain consistency, exchangeability, and integration. To adequately meet the needs of the biology laboratory, the flow cytometer system must meet or exceed the following requirements and specifications. The instrument system must consist of a flow-through particle imaging technology specifically for use in identifying freshwater cyanobacteria and algae from surface water samples collected throughout the state of Florida and shipped to the DEP Laboratory, Tallahassee, FL, instrument control software, and data processing and reporting software. As this will be the first instrument of this kind for the Biology Program, customer training and support must be provided. A market review of flow cytometers for this purpose is also provided.

1. Flow Cytometer Specifications:
1.1 The flow-through, particle digital imaging technology must be a bench top instrument that uses a combination of fluorescence excitation wavelength, auto-imaging, and image recognition software to differentiate cyanobacteria cells from other algae and non-algae particles based on curated taxonomic libraries. Biology is purchasing this instrument specifically for use in identifying cyanobacteria and phytoplankton in freshwater surface water samples. The use of the excitation wavelength to trigger imaging and classification will help delineate similarly shaped cells in different taxonomic groups (for example, to differentiate between cyanobacteria and green algal cells) and assist the user in building robust taxonomic libraries.
1.2 The flow-through, particle imaging technology must be able to measure particle fluorescence and light scattering to discriminate pigmented from non-pigmented particles. The instrument must include a 633 nm laser to trigger auto-imaging and at least two corresponding channels of fluorescence detection (phycocyanin & chlorophyll) to differentiate cyanobacteria from other algal groups and detritus. Phycocyanin, a pigment found in cyanobacteria, excites at 633 nm wavelength whereas chlorophyll-a excites at 488 nm wavelength. Using the phycocyanin excitation wavelength will help differentiate cyanobacteria from other phytoplankton, such as those belonging to Chlorophyceae. DEP must differentiate and identify freshwater cyanobacteria species as part of our interagency harmful bloom response efforts.
1.3 Aquatic objective lens with at least 3 magnifications (4X, 10X, 20X) for counting and imaging particles from approximately 2µm to 600µm is required. Multiple magnification requirements are needed to ensure suitability for the wide range of potential freshwater algal cell, unit and colony sizes present in environmental surface water samples.
1.4 Must operate with minimal additional reagents. When procuring new technologies, Biology must be mindful of budget allocations and add instrumentation that minimizes additional operational costs, staff time to order and stock reagents, and steps that may complicate the analysis.
1.5 Must have multiple settings to optimize the flow rate. Ability to adjust and set the volume of liquid to be analyzed per minute is required. Environmental samples are heterogeneous and flexibility in how much of an environmental sample is analyzed will allow Biology to develop
PUR 7776 (Revised 07/2022) Page 6 of 9 Incorporated by Reference: Rule 60A-1.045, F.A.C.
analytical protocols that meet the data objectives of the wide variety of customers and agency programs.
1.6 The flow-through, particle imaging technology must have adjustments for frame rate and flow rate, so the camera evaluates a different image frame without missing any fluid (misses particles present in the sample) and without overlapping the sample fluid (multi-imaging or double counting). Surface water environmental samples are heterogeneous with many unique algal cells therefore, it is critical that each particle is capable of being imaged and measured individually without bias.
1.7 The instrument must have a high resolution (1920 x 1200 pixels or greater), auto-focus, color camera, with a shutter speed at least up to 60 fps. The ability to obtain clear, color images that support algal cell classification to at least the genus level resolution is required. Color images improve identification capability and capture quality images for use in communication and presentations. This requirement ensures the instrument is comparable to existing laboratory imaging capabilities.
1.8 The instrument must be capable of a sample processing speed of 0.05 mL/minute and up to 3 mL/minute. Operating the instrument under these collection conditions must have negligible impact to data quality and reproducibility. These sample processing speed specifications mimic the rate of current dominant IDs so it will be compatible and ready to integrate with the laboratory’s existing methods.
1.9 The ability to calibrate the instrument on particle size and shape is required. The DEP Laboratory operates using a robust quality assurance program and an accurately calibrated instrument is essential to the mission of producing high quality, repeatable data.
1.10 The ability to run a cleaning cycle between samples to prevent cross-contamination is required. The DEP Laboratory operates using a robust quality assurance program and data results absent of contamination is essential to the mission of producing high quality, repeatable data.
1.11 An instrument footprint that fits on a standard laboratory benchtop, approximately 29 inches in depth. This requirement ensures the instrument is compatible with existing lab space and infrastructure constraints. Ideally, the instrument would have an onboard computer or operating system so that the software and generated raw data is stored on the instrument rather than a separate computer. This is needed to reduce the overall footprint of the instrument as well as the initial and recurring cost of buying or upgrading separate components.
1.12 This instrument must be field-upgradable and compatible with an automated system for sample preprocessing including liquid handling and fully customizable sample analysis. This feature adds additional capability during the operational life of the instrument and positions the Biology Program to meet the longer-term goal of automating aspects of the algal identification workload, specifically for routine samples and specific projects.
2. Data System Specifications:
2.1 A complete, authorized software package for control of the flow cytometer and the output is required. The software should be fully supported by the company without the need for additional third-party software. Software must be Windows® compatible. The software must include the capability to control flow rate, data acquisition, method automation, data analysis and processing, data reporting. Instrument software must be able to export data for long-term storage all category counts and calculated metrics. Export format must be compatible with Microsoft’s Excel spreadsheet application. These software requirements are needed to integrate with the DEP Laboratory Information Management System (LIMS) and Statewide Biological Database (SBIO).
PUR 7776 (Revised 07/2022) Page 7 of 9 Incorporated by Reference: Rule 60A-1.045, F.A.C.
The DEP agency uses Microsoft software and compatibility is ideal for business continuity of operations. When procuring new technologies, Biology must be mindful of budget allocations, agency software licensing policies, and add instrumentation that minimizes additional operational costs, components or software that could compromise or limit business continuity of operations.
2.2 The instrument software must use statistical probabilities to detect, categorize, sort and filter (classify) like-particles by sizes and shapes with exact precision as user-defined criteria set.
Providing a scientifically based, standard, repeatable method for the analytical result (classification) produced is required by the lab QA and for the wide variety of customers and agency programs.
2.3 The instrument must calculate and output a variety of metrics, including but not limited to counts, cell density, and biovolume. The instrument must be able to calculate particle measurements directly from the captured image of individual particle rather overall population distribution. These measurements are needed to enumerate, calculate density, and calculate biovolume of bloom forming cyanobacterial cells. One of the reasons for purchasing an instrument is to achieve quantitative rather than qualitative results. One agency program has requested the lab provide biovolume measurements for their algal samples. Calculation of biovolume by the instrument and software saves precious staff time having to enter taxa lists, retrieve data, develop equations and a storage data base, perform manual calculations, and develop a way to report to customers. Less post-processing translates to cost savings for our customers and enhanced analytical capability.
Instrument should be able to produce the following measurement properties that will be used to generate particle statistics. These properties assist with partitioning and identification of algal cells of similar shape, size, or color in a heterogeneous sample:
• Basic shape and size parameters: Area, Aspect Ratio (width/length), Area Based Diameter (ABD), Equivalent Spherical Diameter (ESD), Length, Volume (ABD-based), Volume (ESD-based), Width, Biovolume Measurements.
• Advanced Morphology Parameters: Area (Filled), Circle Fit, Circularity, Circularity (Hu), Compactness, Convex Perimeter, Convexity, Elongation, Fiber Curl, Fiber Straightness, Geodesic Aspect Ratio, Geodesic Length, Geodesic Thickness, Perimeter, Roughness, Symmetry
• Gray Scale and Color Measurements: Average Blue, Average Green, Average Red, Edge Gradient, Intensity, Blue/Green Ratio, Red/Blue Ratio, Red/Green Ratio, Edge Gradient, Intensity, Sigma Intensity, Sum Intensity, Transparency
2.4 Instrument software must be able to display a collage of images that were categorized together for the user to review. The software must allow the user to compare detected particles from a sample to known taxonomic algal cell reference images and allow the user to manually curate and update the reference libraries and categorization criteria to improve future results. The DEP Laboratory operates using a robust quality assurance program and visually quality checking the data results for taxonomic accuracy is essential to the mission of producing high quality, repeatable data.
2.5 Result exports must be of reasonable file size for storage, for example less than 800 kb.
2.6 The data system must have the ability to process previously acquired and exported data.
This feature is needed for periods of high incoming workload, where staff will have time to run unpreserved samples to capture data yet will have to analyze and perform quality control on the data later.
PUR 7776 (Revised 07/2022) Page 8 of 9 Incorporated by Reference: Rule 60A-1.045, F.A.C.
2.7 Up to 3 software licenses must come with the data system. Multiple licenses will allow flexibility to simultaneously run the samples and analyze results on different computers. The ability to have multiple staff at different levels of capability and expertise process algal samples is one reason for procuring this instrument.
2.8 Example image libraries and taxonomic classification settings must be provided for training and use. Expedites time from instrument receipt to production.
2.9 The software must have an optional e-signature and audit trail tools used to achieve 21 CFR Part 11 compliance.
3. Service, Installation, and Training:
3.1 The Service Provider must include delivery and installation of all equipment, including checkout and verification of performance. The Service Provider must offer timely, on-site services including all labor, parts, and materials required to maintain the hardware in good operating condition. On-site installation provides immediate verification that the instrument is working properly before the laboratory officially accepts the purchased equipment. This instrument will be used for harmful algal bloom response which requires same day or next day results. When procuring new technologies, Biology must be mindful of budget allocations and add instrumentation with services that minimizes maintenance costs and downtime that could compromise or limit business continuity of operations.
3.2 The Service Provider must be the original equipment manufacturer (OEM) or a certified manufacturer distributor. The equipment must not be modified against the original manufacturer’s recommendation. This ensures that the instrument does not void the OEM warranty and that service and parts will be available during the life of the instrument.
3.3 The flow cytometer Data Acquisition System must include maintenance warranty and technical support of at least one (1) year including all parts, software upgrades, labor and travel, and have available optional enhanced/extended warranty packages. Warranty service is needed to mitigate potential instrument failure and reduce instrument down-time.
3.4 The vendor must provide ongoing service agreements using OEM-trained service engineers for the entire flow cytometry system, including the instrument, computer, and data collection system.
3.5 The Service Provider must include instrument familiarization/training at the time of instrument installation or at a mutually agreed time following instrument installation.
3.6 The Service Provider must provide access to on-demand online training. This helps ensure the laboratory can receive quality service during start-up and over the life of the instrument.
3.7 The service provider should be able to deliver the instrument and provide verification that the instrument is working properly by the state’s 2022/2023 fiscal year deadline of September 30, 2023. Biology must verify that the instrument is working properly to officially accept the purchased equipment before final payment can be issued.
