About this file

This document is a Description of Intended Single Source Purchase (PUR 7776) issued by the Florida Fish and Wildlife Conservation Commission (FWC) for a KASP Genotyping Assay Development for Corals. The intended use of the KASP genotyping assays is to genotype the over 3,000 corals collected as part of the Coral Rescue Project, a joint effort between FWC and NOAA, to preserve genetic diversity and propagate them for restoration of Florida's Coral Reef. The contract term is three (3) calendar years from issuance of the first purchase order. Vendors must provide information regarding their ability to supply the services by April 26, 2024 at 10:30 AM ET.

The estimated dollar amount for the contract is $275,000, which includes $16,000 per species for 16 species with an additional $3,000 buffer. FWC has identified LGC Biosearch Technologies as the sole source provider for the KASP assays, as they are the patent holder and manufacturer of the required products and services, which have an industry-leading accuracy rate of over 99.8%.

This description of commodities or contractual services intended for purchase from a single source is posted in accordance with Sections 120.57(3) and 287.057(3), Florida Statutes (F.S.), and will remain posted for a period of at least 15 business days.

Commodity or Contractual Service Required (commodity or United National Standard Products and Services Code (UNSPSC), manufacturer, model, and description, as appropriate):

85131709: Genetics research services

Quantity or Term (as appropriate):

Three (3) Calendar Years from issuance of 1st purchase order.

Requestor (division, bureau, office, individual, as appropriate):

Alicia Vollmer

Performance and/or Design Requirements (e.g. intended use, function or application, compatibility, requirements; reference to policy, rule, statute or other act of the Legislature, etc., as appropriate):

The intended use of the Kompetitive Allele Specific PCR (KASP™) genotyping assays is to genotype the corals collected as part of the Coral Rescue Project. Over 3,000 corals were collected from the wild as part of the Coral Rescue Project, a joint effort between FWC and

NOAA, and placed in land-based facilities to preserve genetic diversity and propagate them for restoration of Florida's Coral Reef in response to stony coral tissue loss disease. All of the corals in holding are the property of FWC and are on loan to authorized aquariums around the US. All of the corals in holding need to be genotyped to identify unqiue individuals for breeding and propagation. These KASP genotyping assays use 192 species-specific single nucleotide polymorphism (SNP) markers discovered during the genotyping-by-sequencing process to

develop a genotyping assay in which coral samples are analyzed. Once the assay is created for each coral species, it can be repeatedly used to genotype additional corals that are collected, identify the genetic diversity of baby corals that are propogated in hodling facilities or examine the relatedness of outplanted populations at restoration sites. The genetic infromation from the

KASP assay (and the genotyping data) is then entered into the FWC Genetic Registry, which houses all of the raw genetic data and allows staff to create genetically informed breeding and restortaion plans for the corals.

Intended Single Source:

LGC Biosearch Technologies

Estimated Dollar Amount:

$275,000.00 (~$16,000 per species) x 16 species w/ additional $3,000.00 buffer.

Justification for single source acquisition (what is necessary and unique about the product, service or source; steps taken to confirm unavailability of competition, as appropriate):

Kompetitive Allele Specific PCR (KASP™) assays enable accurate, bi-allelic discrimination of single nucleotide polymorphism (SNPs) markers. This is a proven method for genotyping corals within the Coral Rescue Project as we have created KASP assays for 5 coral species already. We have attempted other techniques for creating genotyping assays that can be used for corals, but they have failed and provide unusable results. The KASP assay method has an accuracy rate of

>99.8% based on an independent assessment and is an industry leading SNP conversion rate. The

KASP-TF Master Mixes, KASP Assays (KASP By Design, and KASP On Demand) and KASP

All-Inclusive genotyping services are sole source products and services manufactured and operated by the LGC Biosearch Technologies, and for which the company own the patent rights.
