INTRODUCTION

THIS IS A PRE-SOLICITATION NON-COMPETITIVE (NOTICE OF INTENT) SYNOPSIS TO AWARD A

CONTRACT OR PURCHASE ORDER WITHOUT PROVIDING FOR FULL OR OPEN COMPETITION.

The National Institutes of Health, National Institute of Child Health and Development (NICHD) intends to award a purchase order without providing for full and open competition to Cyagen US INC., 2255 Martin Avenue, Suite E, Santa Clara, CA. 95050, USA, for generation of conditional knock-in/out mouse lines.

Our research focuses on the development of the mammalian hematopoietic system. A long-term area of interest for our laboratory is the study of signal transduction molecules and pathways that regulate T lymphocyte maturation in the thymus. Currently, we are analyzing transgenic and conditional nonsignaling mutants of T cell antigen receptor (TCR) signaling subunits to evaluate the importance of individual TCR signaling chains and motifs at specific stages of T cell development and in mature T cells. We are also using gene profiling to identify proteins that are important for fine-tuning the T cell–signaling response in developing and mature T lymphocytes. In conjunction with checkpoint inhibitors for immunotherapy in humans, these molecules may also be targets. We are also investigating the function of new T cell–specific proteins that we identified by subtraction library screening. We have identified two gene candidates: LAX1 and SIT1, that we would like to make conditional knockin/knockout mouse models of to future investigate. LAX1 and SIT1 are involved in the T cell receptor pathways and are genes that are related to our ongoing Themis 1 & 2 and 6Y/6F projects in the lab.

Cyagen offers a proprietary service called TurboKnockout, which allows for the generation of conditional knockin/knockout mouse models though ES targeting cell lines in 6-8 months instead of the conventional 10-14 months. They are the only company that offers the TurboKnockout ES cell targeting conditional knock-in/out mouse model generation service. They have a “super” competent ES cell line that generations 100% ESC-derived founder mice rather than chimeras, eliminating the need to screen for germline transmission. The company also has a selection cassette on the targeting construct capable of removing itself after ES cell targeting without the need to breed to Flp delete mice. Both of these proprietary steps eliminate the need for two generations of breeding and shortens the production time by 4-6 months as compared to conventional standards, which has more steps that require more time. The shortened time would greatly benefit our program goals.

Before awarding a contract to an offeror, the organization shall file, with the Office of Laboratory Animal Welfare (OLAW), National Institutes of Health (NIH), a written Animal Welfare Assurance (Assurance) which commits the organization to comply with the provisions of the PHS Policy, the Animal Welfare Act, and the Guide for the Care and Use of Laboratory Animals (National Academy Press, Washington, DC). In accordance with the PHS Policy, offerors must establish an Institutional Animal Care and Use Committee (IACUC), qualified through the experience and expertise of its members, to oversee the institution’s animal program, facilities, and procedures.

Offerors must provide verification of IACUC approval prior to receiving an award involving live vertebrate animals. No award involving the use of animals shall be made unless OLAW approves the Assurance and verification of IACUC approval for the proposed animal activities has been provided to the Contracting Officer.
