About this file
This document is a justification for other than full and open competition at or below the simplified acquisition threshold. The National Institute of Allergy and Infectious Diseases Vaccine Research Center seeks to procure a Wyatt miniDAWN instrument with accessories, software, installation, training and support in order to characterize macromolecules and nanoparticles for the detection of antibodies directed against respiratory virus spikes and glycoproteins. The Vaccine Research Center identified this equipment as the most reliable and sensitive system over the last 10 years that offers high dynamic range and throughput for accurate measurement of molar mass and root mean square radius, critical for developing viral proteins to detect antibodies and inform vaccine design and modification against emerging respiratory viruses such as COVID-19.

Justification for Other than Full and Open Competition:
at or Below the Simplified Acquisition Threshold
I. Identification of the agency and contracting activity
| DHHS, NIH, NIAID, VRC, PREMISE | |
| Address of the requesting lab/office: | 9 West Watkins Mill Road Gaithersburg, 20878 |
A. Project Background
a. The PREMISE program aids the Vaccine Research Center (VRC) by supporting the development of monoclonal antibodies and vaccines for infectious diseases such as HIV, Influenza, Ebola and other respitoray pathogens such as Covid and parainfluenza virus. Support is through the state-of-the-art laboratory performing Phase 1 and Phase 3 clinical end point testing to inform further study designs, research questions and vaccine modification and design. As part of a broader strategy to accelerate the development of monoclonal antibodies as therapy for targeting respiratory viruses and newly emerging variants, VIP has identified the electrochemiluminescent (ECL) assay format as the most reliable and sensitive assay system that offers high dynamic range in the detection of antibodies isolated specific viral entry spikes and glycoproteins.
b. Acquisition purpose and objectives.
The purpose of the procurement is for the vendor to provide the Wyatt miniDAWN and Optilab equipment that we will use to synthesize proteins.
II. Identification of the statutory authority permitting other than full and open competition (SELECT ONE)
☒ 41 USC 3304(a)(1) and FAR Part 6.302-1: Only one responsible source/Brand Name
The threat of emerging respiratory viruses continues in the international community, the need for a safe and efficacious monoclonal antibody therapy at an accelerated timeline is a health care priority. To assist in this effort, the PREMISE will use the Wyatt MiniDAWN to characterize macromolecules and nanoparticles which we have developed for the detection of specific antibodies directed towards a panel of respiratory virus spikes/glycoproteins. These proteins will then be 'printed' or 'spotted' in a single well of a 96-well microtiter plate. PREMISE identified this equipment as the most reliable and sensitive system over the last 10 years, that offers high dynamic range in the detection of specific particles and serum antibodies that will be used to develop a variety of viral proteins. The platform offers extremely high throughput and and includes 3-angles of scattering detection for accurate measurements of molar mass (Mw) and root mean square radius (Rg).
