About this file
This Statement of Work (SOW) outlines genetic analysis requirements for the US Geological Survey's monitoring of juvenile fall Chinook salmon in Snake River. USGS will submit 980 tissue samples preserved in 90% ethanol or on Whatman paper, along with corresponding sample numbers, fish measurements, and sampling dates. The vendor must perform SNP genotyping and parentage analysis using a minimum of 192 SNP genetic markers to validate fish origin estimates through parentage-based tagging (PBT) and genetic stock identification (GSI). The period of performance is December 23, 2024 through May 31, 2025.
The associated Notice of Intent indicates USGS plans to award a sole-source purchase order to Idaho Fish and Game's Eagle Fish Genetics Lab, identified as the only source conducting annual PBT analyses on all fall Chinook salmon in hatcheries. Responses are due by December 20, 2024 at 0800 hours central time via email to rlujan@usgs.gov. The opportunity falls under NAICS code 541380 (Testing Laboratories and Services) and PSC B516 (Special Studies/Analysis Animal/Fisheries).

Statement of Work Parentage-Based Tagging Analysis
The USGS has long-term monitoring projects in place on Snake River to estimate abundance of juvenile fall Chinook salmon migrating through Lower Reservoir. As part of this monitoring, the USGS estimates the origin (i.e., fall versus spring/summer) of the fish they sample; however, these estimates can only be validated with genetic analyses. The USGS will validate their origin estimates of fish sampled during 2024 using genetic screening and parentage-based tagging (PBT)/genetic stock Identification (GSI) analyses.
The USGS will prepare and submit 980 samples according to procedures specified by the vendor, which is preservation of tissue in 90% ethanol or dry storage on Whatman paper. The USGS will also supply a list of sample numbers, fish length and weight, and sampling date for each sample.
The vendor will analyze tissue samples from sampled fish using SNP genotyping and parentage analysis. Samples will be screened with a minimum of 192 SNP genetic markers. This methodology is currently used to type all hatchery fall chinook salmon in the Snake River basin. Thew vendor will provide the results for each sample in an Excel file summarizing PBT/GSI assignments and genetic sex.
Period of Performance (estimate): 12/23/2024 – 05/31/2025
