About this file

This document is a Statement of Need (SON) for the National Marine Fisheries Service's Genetics & Evolution Program within the Conservation Biology Division at the Northwest Fisheries Science Center (NWFSC). The SON outlines the requirement for a Thermo Scientific KingFisher Apex purification system to support the program's advanced genetic and genomic research on marine species conservation.

The key details include: the KingFisher Apex must be a programmable, magnetic bead-based system with customizable protocols, able to process up to 96 samples per run, and have integrated heating/cooling, flexible software, and multiple magnetic head options. It must be compatible with existing protocols, prevent cross-sample contamination, and include a warranty and service contract. Thermo Fisher Scientific is the sole provider of the KingFisher Apex system and the only vendor capable of meeting the NWFSC's requirements. This SON is related to a Special Notice for a sole-source procurement of the KingFisher Apex system from Thermo Fisher Scientific.

STATEMENT OF NEED

Background

The Genetics & Evolution (GE) Program within the Conservation Biology (CB) Division at the Northwest Fisheries Science Center (NWFSC) conducts science to support the conservation of marine species and the ecosystems upon which they depend - primarily off the coasts of Washington and Oregon and in freshwater rivers and streams in Washington, Oregon, Idaho, and Montana. The primary research activities included in the GE Program portfolio include using advanced genetic and genomic techniques to assess genetic variation in aquatic animals, identify the composition of mixed fishery stocks, estimate fitness in the wild, and evaluate effects of hatchery domestication, fishing, and climate change. The GE Program also uses environmental DNA (eDNA) to monitor diversity, distribution, and abundance of marine species in open ocean and nearshore communities.

NWFSC’s Genetics and Evolution Program requires a Life Technologies Corporation dba Thermo Scientific KingFisher Apex purification system to support the high throughput NGS genomics needs of NWFSC’s Molecular Genetics Laboratory. NWFSC’s laboratory employs a wide array of molecular genetic approaches to support a diverse project portfolio to meet NOAA missions and requires an instrument that supports the flexibility needed to purify DNA/RNA from a variety of custom and commercially available extraction protocols to further support the needs for high throughput sample processing. This system is also needed to further reduce (eliminate) cross-sample contamination and additionally support molecular geneticists by reducing the potential physical effects of repetitive strain injuries associated with manual sample processing. This instrument is needed to support both ongoing work and projects directly supported by Inflation Reduction Act funding. The automated system must be compatible with existing validated protocols and reagents and must include a warranty and service contract to avoid interruption of service.

General Requirements:

1. Magnetic bead-based programmable system for purification of DNA/RNA that supports customizable protocols consistent with in-house purification protocols and commercially available extraction kits from a variety of manufacturers.

· An open system to allow use of user selected magnetic particle bead reagents.

2. Medium to high throughput automated system consisting of KingFisher magnetic particle purification instrument, optimized DNA/RNA purification kits, plastic consumables and BindIx Software.

3. Instrument can be used for nucleic acid, protein and cell purification applications

4. Bench top instrument with footprint not to exceed (99 x 77.5 x 83.5 cm), weight 58 kg 5.Process up to 96 samples per run with total working volume from 20 μl to 5000 μl.

6. User interchangeable magnetic head with 4 different options o 96DW magnetic head for 96 samples for volumes 20-1000 μl o 24DW magnetic head for 24 samples for volumes 200- 5000 μl o 96KF magnetic head for 96 samples for volumes 20-200 μl o 96PCR magnetic head for 96 samples for volumes 20- 100 μl

7. Instrument uses permanent earth magnetic rods covered with sterile, single use, disposable plastic tip “combs” during processing to ensure protection of samples and prevent sample contamination.

7. A flexible software system to allow ‘on the fly’ customer protocol modifications that is compatible with reagents from other (non-Eppendorf) manufacturers (e.g. Qiagen, ZymoResearch).

8. Integrated heating and cooling capability for all magnetic head types o Heating/cooling capability from +4°C to +100°C

9. Shield plate which moves under magnetic head during rotation of the round table

10. Fast protocol run times, typically 30-60 min depending on the applications.

11. One year manufacturer’s warranty inclusive of coverage for defects in workmanship and materials that covers all parts and labor.

12. A service agreement (optional years) that covers the instrument after the warranty period, includes an annual preventative maintenance calibration service performed by OEM-certified service professionals, priority access to technical support (Monday-Friday), for 4x basic dispensing tools, and includes annual preventive maintenance onsite visit, an additional repair visit once per year inclusive of costs associated with travel, labor and parts and software updates during the warranty and service periods.

Delivery shall be made to:
