About this file
This is a Minimum Requirements document for an automated inverted microscope system acquisition by the National Institute of Standards and Technology (NIST). NIST's Biophysical and Biomedical Measurement Group seeks a complete microscope system to support its Organ-on-a-Chip and Microphysiological Systems research program, with emphasis on live cell imaging capabilities and compatibility with existing Zeiss microscope infrastructure in the laboratory.
The system comprises eight line items with detailed technical specifications: (1) an inverted microscope body with motorized focus drive (20 nm minimum step width), motorized nosepiece, filter turret, and motorized X-Y scanning stage (100 mm travel range, 0.5 mm resolution) capable of bright field and reflection imaging plus phase contrast and differential interference contrast microscopy; (2) four microscope objectives ranging from 2x to 20x magnification with specified numerical aperture and working distance requirements; (3) a CMOS camera with greater than 15 megapixels resolution, forced-air cooling to 25°C, USB 3 and PCIe interfaces, 20+ fps frame rate, C-mount lens mount, and 1-16x signal amplification; (4) a multi-channel LED illumination source with integrated control unit and minimum 4-channel fluorescent capability (7-channel preferred) covering UV through far-red wavelengths (385-735 nm) with filter sets for standard fluorophores; (5) microscope control software with graphical user interface supporting image acquisition, processing, automation of motorized components, time-lapse imaging, z-stack capability, and compatibility with Zeiss software; (6) heating and temperature control unit (37°C for mammalian cultures) and CO2 control unit; (7) a workstation-class personal computer with Intel Xeon processor, 32 GB RAM minimum, dedicated Nvidia graphics card with 16 GB memory, 2 TB SSD storage, and 30-inch monitor; and (8) interchangeability of parts and software with existing Zeiss microscopes. All equipment must be new, manufacturer-packaged, and include original documentation and software.
