ISSUED BY: National Institutes of Health (NIH), National Cancer Institute (NCI), Office of

Acquisitions (OA), Strategic acquisitions Branch (SAB)

DESCRIPTION:

The Department of Health and Human Services (HHS), National Institutes of Health (NIH), National Cancer Institute (NCI), Center for Cancer Research (CCR), Laboratory of Cellular

Oncology (LCO) has a need for overlapping peptides of the rabies envelope glycoprotein and

HPV capsid L1 protein and antigen.

This is a Combined Synopsis/Solicitation for commercial items, prepared in accordance with the format in Federal Acquisition Regulation (FAR) subpart 12.6, as supplemented with additional information included in this notice, and shall be processed in accordance with FAR Part 13–

Simplified Acquisition Procedures. This announcement constitutes the only solicitation that will be provided in relation to the stated need contained herein. This solicitation is issued as a

Request for Quote (RFQ) and includes applicable provisions and clauses in effect through FAR

Federal Acquisition Circular (FAC) 2023-04 (06/02/2023).

The requirement’s North American Industry Classification System (NAICS) code is 325413

(IN-VITRO DIAGNOSTIC SUBSTANCE MANUFACTURING) for which the size standard is

1,250 Employees.

One Firm-Fixed-Priced award is anticipated to be made as a result of this solicitation.

1.0 BACKGROUND

The Laboratory of Cellular Oncology aims to evaluate the biologic and therapeutic potential of overlapping Viral Peptide Libraries (VPLs). The overall goal is to develop broadly applicable biological therapies for cancer with a product profile that is suitable for use in low resource settings. Using this approach, the laboratory seeks to address the vast disparities in access to cancer control measures in high- and low-income settings that is being exacerbated by the increasing focus of cancer research community on personalized approaches to treatment.

To implement these studies, the Laboratory of Cellular Oncology requires the design, synthesis, pooling, purification, and characterization of a VPL of overlapping sequences, derived from the major antigens present in Human Papillomavirus (HPV) 16 viral capsid protein L1 (HPV16-vcpL1), present in the HPV vaccine Gardasil and of rabies virus envelope glycoprotein (Rabies-gpENV), present in the inactivated rabies vaccines. The laboratory will use these VPLs to determine tolerability, immunological response, and anti-tumor efficacy; to assess the safety and effectiveness of such library, and to complement currently open human trials designed to test new doses and effects of this agent.

2.0 SCOPE

The Contractor shall provide all labor materials, and facilities required to provide services in the area of peptide design and synthesis.

Based on the entire amino acid sequence of the HPV16-vcpL1 and Rabies-gpENV, the

Contractor shall generate a list of peptide sequences, with 124 sequences in total for HPV16-vcpL1 and 129 sequences in total for Rabies-gpENV. These peptide sequences shall be of 15 amino acid residues in length (15-mers), with an overlap of 11 residues. Based on this list, the

Contractor shall synthesize, purify, analyze, pool, and test the peptides. The Contractor shall then deliver aliquots of pooled peptides and individual peptides to the Government of 15 amino acid residues and 11 residues overlap to cover the entire sequence of HPV viral capsid protein L1 and

Rabies envelope glycoprotein to capture all MHC-1 and MHC-2 restricted epitopes. The target amount of peptide shall be a minimum10MG.

3.0 SPECIAL ORDER REQUIREMENTS

The Contractor shall provide the following services:

3.1 Peptide library design:
