About this file

This request for quotation (RFQ) from the National Cancer Institute (NCI), a division of the National Institutes of Health (NIH), solicits quotes for the procurement of one Nikon A1R HD25 confocal system with a Ti2-E inverted microscope. The Laboratory of Cell Biology (LCB) at NCI's Center for Cancer Research seeks to increase the throughput of its zebrafish model for studying cancer metastasis by acquiring a confocal system and microscope with a larger field of view and higher spatial resolution than its current equipment. The required product features for the microscope include a 25mm field of view, offset lens for focus correction, built-in DAQ card, TIRF and confocal microscopy capabilities, and specified objective lenses. The AIR HD25 confocal scan head must provide seamless transition between galvano and resonant scanners, 1K x 1K spectral detection across the field of view, and resonant scanning speeds of 60 fps at 256 x 256 pixels or 15 fps at 1024 x 1024 pixels. The NIS-Elements C software is also required and must minimize noise during scanner switching and remove Poisson detector shot noise. Quotes are due by the date specified in the RFQ.

REQUEST FOR QUOTATION

75N91021Q00022

1.0 TITLE

Procurement of one (1) Brand-Name or Equal NIKON A1R HD25 Confocal System with Ti2-E Inverted Microscope.

2.0 BACKGROUND

The National Institutes of Health (NIH), National Cancer Institute (NCI), Center for Cancer Research (CCR), Laboratory of Cell Biology (LCB) is focused on research that studies what drives selection of the sites of metastasis during cancer progression. Specifically, longitudinal imaging using a novel human xenograft in zebrafish metastasis model is key to understanding this complex question. These sites of metastasis are non-random; for example, breast cancers preferentially grow in lungs bones and brain whereas uveal melanoma preferentially grow in the liver. Two hypotheses have been put forward to address this non-randomness: 1) seed/soil hypothesis: metastasis sites are largely linked to the biochemical compatibility between cells and the secondary organ environment and 2) mechanical hypothesis: the non-random behavior is driven by the architecture and fluid dynamics of the interconnected vessels. The LCB wishes to visualize these early steps of organ colonization under multiple perturbations in order to begin to determine what governs organ selectivity of metastasizing tumor cells in vivo.

To accomplish this, the LCB seeks to increase the throughput of its zebrafish model, allowing researchers in the lab to rapidly image the entire fish without compromising spatial resolution and to examine sub-cellular protrusions in multiple organs with near simultaneity. The LCB currently images multiple fixed and live sample types including zebrafish, organoids, and tissue explants. The systems currently used by the lab are too slow to acquire the necessary field of view and do not possess the spatial resolution needed to further the research conducted by the LCB. Thus, the LCB seeks to procure a new confocal system and inverted microscope with a larger field of view and higher spatial resolution.

3.0 PRODUCT FEATURES/SALIENT CHARACTERISTICS

The following salient characteristics are applicable to this acquisition.

3.1 Ti2-E Microscope

3.1.1 Shall include a large field of view/field of vision (FOV) of at least 25-mm, coupled with a dual-camera support (64 x 64 through 4096 x 4096).

3.1.2 To ensure that there is no large drift due to this large field of view, an offset lens shall be included that will allow simultaneously monitoring of the reference focal plane and maintaining focus on the image plane in real-time with continuous correction.

3.1.3 Shall include a built-in DAQ card that allows the end-user to add an external trigger or sync their image acquisition with an additional modality

3.1.4 The microscope shall have the capability to move between total internal reflection microscopy (TIRF) and confocal microscopy using motorized parts and the built-in software.

3.1.5 Shall include the following laser lines:

3.1.5.1 405

3.1.5.2 408

3.1.5.3 561

3.1.5.4 640

3.1.6 Shall be capable of the following objective magnifications:

3.1.6.1 Plan Apo Lambda 22X, NA 0.1, WD 8.5 mm

3.1.6.2 Plan Apo Lambda 10X, NA 0.45, WD 4.0 mm

National Cancer Institute 75N91021Q00022 Page 2 of 24

3.1.6.3 Plan Apo Lambda 20X, NA 0.75, WD 1.0 mm

3.1.6.4 Plan Apo Lambda 60X Oil immersion NA 1.4, WD 0.13 mm

3.1.6.5 Apo Lambda LWD 20X Water immersion lens NA 0.95, WD 0.95mm

3.2 AIR HD25 Confocal Scan Head

3.2.1 To achieve temporal resolutions spanning milliseconds to seconds, two types of scanners are typically done for each speed. The scan head shall ensure a seamless transition between a Galvano-scanner and resonant scanner and allow for switching between each scanner without restarting the software.

3.2.2 The scan head also shall have high resolution spectral detection of “1K X 1K” for the entire field of view that does not require scan field zooming or cropping. Thus, reduced image resolution is not lost as is usually the case when imaging a large field of vision at higher temporal speeds.

3.2.3 The resonant scanner shall allow imaging at a minimum acquisition speed of 60 frames/s (256 x 256 pixels), 30 frames/s (512 x 512 pixels), or 15 frames/s (1024 x 1024 pixels) at 1x optical zoom or line scans at rates of 720 frames/s at 512 x 16 pixels and up to 15,600 lines/s.

3.2.4 The Galvano scanner shall have a 25 mm diagonal FOV with continuous zoom option from 1-1000x.

3.2.5 The pinhole shall allow for optimized light transmittal in both the excitation and detection of light from one imaging plane, and thus must be a geometric circle or hexagonal shape.

3.3 NIS-Elements C Software

3.3.1 Software shall be able to trigger switching from different scanner without restarting and must have a function to minimize noise due to switching between instrument heads.

3.3.2 Software shall include the ability to remove Poisson detector shot noise from confocal images, significantly reducing the need for image averaging, and allowing users to push their imaging to higher speeds at lower laser powers ( <1% of 1mW). This ability should be able to be applied live or post- image acquisition.

3.3.3 Software shall have a function for an integrated water dispenser for adding and removing water during time-lapse and multi-position experiments using software for full experimental control.

3.3.4 Software shall have an AI-based image analysis for label-free segmentation and thresholding of images.

3.4 TRAINING
