THIS SOLICITATION IS A SOLE SOURCE REQUIREMENT PRAESENBIO, LLC ONLY.

1. NOTICE TO ALL OFFERORS: The Government anticipates awarding a firm, fixed price contract in response to quotes/proposals received to this solicitation. As a result, the contract's pricing will not be subject to any adjustment on the basis of the contractor's increased costs due to COVID-19 and/or inflationary causes experienced during contract performance.

Because a firm, fixed price contract places the maximum risk and full responsibility for all costs and resulting profit or loss on the contractor, offerors are advised to consider these risks when pricing their quotes/proposals.

2. This solicitation is for the procurement of Sequence-Based Detection in support of US Naval Medical Research Unit TWO (NAMRU-2). It is anticipated that this solicitation will result in an award of a Firm, Fixed-Price Service type contract.

3. The entire solicitation shall be filled out, signed, and returned as part of your quote. See block 8 on page 1 of the solicitation for the closing date and time.

4. The deadline for submission of questions regarding the solicitation’s terms, conditions, specifications, and/or the statement of work (SOW) / performance work statement (PWS) is 4 working days prior to the solicitation’s closing date.

5. The Government will award on the basis of price unless the contracting officer is aware of past performance information related to the low price quote/offer which indicates that quote/offer may not represent best value. In that case, the Government reserves the right to consider the past performance of other quotes/offers, conduct a price, past performance tradeoff, and award to other than the lowest price quote/offer.

6. SINGLE AWARD FOR ALL ITEMS: Due to the interrelationship of supplies and/or services to be provided hereunder, the Government reserves the right to make a single award to the quoter whose quote is considered in the best interest of the government, price and other factors considered. Therefore, quoters who quote less than the entire effort specified herein may be determined to be unacceptable.

FOR

SEQUENCE-BASED DETECTION AND CHARACTERIZATION OF EXISTING AND EMERGING

BIOTHREAT AGENTS, ARBOVIRUSES, AND ANTIMICROBIAL RESISTANCE GENES OF

MULTIDRUG RESISTANT MICROBIAL PATHOGENS IN SOUTH-EAST ASIA AND OCEANIA.

1. OBJECTIVE

1.1 The objective of this contract is to obtain contractor support services for the execution of detailed molecular sequencing activities to support NAMRU-2’s surveillance activities focused on microbial pathogens endemic in the Indo-Pacific region, to encompass current and novel biothreat/bioterrorism agents and antimicrobial resistant microorganisms, as well as arboviral diseases of consequence to the warfighter.

2. INTRODUCTION

2.1 The mission of the U.S. Naval Medical Research Unit TWO (NAMRU-2) is to monitor and characterize emerging and re-emerging infectious diseases of military and public health importance, and to develop mitigation strategies against these threats in partnership with host nation, international, and U.S. agencies in Indo-Pacific region. To accomplish this mission, NAMRU-2 contracts with the private sector for management, administration and labor for operations and field research. The contractor will follow an approved protocol for analyzing biological specimens for research and surveillance purposes to understand the infectious disease threats for deployed military personnel and the local population.

2.2 The primary, overarching objective of this project is to support existing health facility and research based infectious disease surveillance networks that currently surveille for and identify biothreat and AMR/MRDO illness etiologies among patients and other volunteer subjects, as well as zoonotic and environmental sources in various countries around the Indo-Pacific Command (INDOPACOM) Area of Responsibility (AOR). The specific objectives to be met by this contract are to support existing NAMRU-2 biosurveillance projects across INDOPACOM AOR by the following:

2.2.1 implement standardized laboratory procedures to detect biothreat agents, arboviruses and antibiotic resistant organisms from contrived, environmental, zoonotic or patient samples,

2.2.2 develop detailed consensus genomes for a select subset of regional pathogens of concern,

2.2.3 provide the capability to identify novel emerging and re-emerging infectious diseases in the region.

2.3 The Contractor shall provide laboratory, training, and logistical support services relating to molecular sequencing support for conducting the NAMRU-2 research programs under their respective IRB protocols, or the equivalent locally approved and administered IRB.

3. SCOPE

3.1 Contractor support shall be required for coordination of activities described in this PWS. Efforts include facilitation of agreement between collaborators, supporting milestone completion, ensuring compliance with research protocols, and maintaining open communication with associated project collaborators and investigators.

3.2 This work shall take place at laboratory sites in the Indo-Pacific region where NAMRU-2 bio-surveillance and research projects are taking place. The work shall include collecting and testing samples by preliminary analyses. Previously collected and stored samples will be analyzed by molecular sequencing and downstream bioinformatics analysis (computer based).

3.3 The contractor shall provide progress reports on the status of the study, as well as monthly telephone calls or virtual meetings to review progress with the NAMRU-2 Principal Investigator (PI) and Co-Investigators, as needed.

4. BACKGROUND

4.1 Emerging and re-emerging diseases throughout the Indo-Pacific region continue to be of great concern during this time of rapid population growth, increasing urbanization, extensive air travel, environmental changes, and the lack of effective intervention strategies. It is of vital importance to understand pathogen circulation throughout these regions and populations and potential dissemination to other regions of the world. Biothreat agents have great harm potential to the warfighter, especially when syndrome-based diagnostics can significantly delay correct etiology identification. As such, there is an urgent need to establish and maintain a surveillance network in strategic locations that will lead to early recognition of emerging pathogens to implement intervention measures to minimize the impact on human health.

4.2 PraesensBio, LLC, Lincoln, Nebraska, is uniquely positioned to serve as the implementing contractor because: (1) it holds a proprietary methodology for sequencing that is low-cost and pathogen agnostic by pathogen- specific enrichment; (2) has the expertise and logistics capabilities needed to train the local partners at the reference laboratories and conduct the detailed genomics laboratory analyses, and (3) has the specialized bioinformatics resources needed to finalize the characterization and description of the genomics data resulting from the analyses. An additional unique and critical component of the PraesensBio, LLC, technology is that the methodology can be applied to diverse on-site equipment types from various manufacturers as may be found within the various NAMRU-2 biosurveillance partners across the region.

5. SPECIFIC TASKS

5.1 The contractor shall:

5.1.1 Bear the primary responsibility for the work.

5.1.2 Implement support for NAMRU-2 studies under their respective ethics protocol, as pertinent, or the equivalent locally approved protocol, hereinafter referred to as the “protocol.”

5.1.3 In collaboration with NAMRU-2, provide managerial, supervisory, administrative, and logistical support to execute activities. In addition, provide the necessary personnel, materials, equipment, facilities, licenses - and all other direct and indirect costs associated with this project.

5.1.4 This requirement involves extramural human subject research; therefore, in addition to the requirements set forth in the basic contract, prior to the Government obligating funding that supports human subject research, provide to the NAMRU-2 Principal Investigator (PI), and make available to the Contracting Officer if requested, the appropriate human use approvals (e.g., Medical Research and Ethics Committee approvals) and the NAMRU-2 Human Research Protection Program (HRPP) review will be retained for review by the NAVSUP Fleet Logistics Center (FLC), Puget Sound Contracting Officer, upon request. The human research approval requires prior review by the in-country partner and NAMRU-2. In the case of an existing local and USN HRPO protocol for regional sites, the contractor shall support any modification to the protocol as may be required. The anticipated sites are (1) the Central Public Health Laboratory and Institute for Medical Research laboratory in Port Moresby and Goroka, Papua New Guinea, (2) the Laos Oxford Mahosot Wellcome-trust Research Unit in Vientiane, Laos, (3) Malaysian Armed Forces sites in Selangor, Kedah, Johor, and Sabah, (4) University of Malaya-Sabah clinics/sites,

(5) the National Institute for Hygiene and Epidemiology in Hanoi, Vietnam, and (6) the 108 Military Central Laboratory in Hanoi, Vietnam.

5.1.5 Ensure that prime and subcontractors providing human subject research shall have and maintain throughout this human research effort an active Federal Wide Assurance (FWA) if the activities are conducted as human subject research.

5.1.6 Provide analysis of the local capabilities to implement and execute sequencing of target samples and expert consultation on the implementation and execution of sequencing support for the studies.

5.1.7 Ensure that staff supported by this contract are trained to handle and process, blood, respiratory, or other types of specimens, including collection, preparation, safe receipt, transport, and storage of potentially infectious samples if needed.

5.1.8 Identify, advise on collection of suitable samples for analysis, analyze and maintain data of all samples according to study protocol, in coordination with instructions from NAMRU-2 and local partners, ensuring local safety requirements and established standard operating procedures are adhered to.

5.1.9 Coordinate with NAMRU-2 and local partners to ensure import and custom clearance procedures for shipping and transport.

5.1.10 Coordinate with NAMRU-2 and local partners to ensure access to appropriate storage, transportation, and shipping of samples and/or reagents (e.g., shipper boxes, - 70 to -80oC in freezers, etc. with constant battery/generator backup in addition to regular outlet electrical supply, other).

5.1.11 Provide summary reports highlighting force health protection relevant research findings. Provide one final project report covering project activities and findings.

5.1.12 Ensure that NAMRU-2 is informed as soon as feasible, should specific project goals not be achieved or problems are encountered.

5.1.13 In collaboration with NAMRU-2, complete the following non-human subject research activities:

5.1.13.1 Oversee on-site training of staff at NAMRU-2’s collaborating sites (at two sites), to (1) process sequencing methodologies via a contamination prevention and control strategy to include genetic barcoding procedures with good clinical practices (GCPs) and quality control procedures, (2) conduct custom hybridization-enrichment, amplicon-based, and metagenomics MinION and/or Illumina-based sequencing on site, as appropriate for the site, (3) and analyze genomics data from sequencing by bioinformatics that can identify, classify and define known and new mutations in known pathogens or characterize near- neighbor novel emerging pathogens. When on-site training is not possible, virtual training and consultation with local partners shall be provided by the Contractor.

5.1.13.2 Provide an integrated system for molecular sequencing at each laboratory to include (1) modified molecular sequencing workflow SOPs tailored for each site’s specific laboratory setup as needed, (2) sample processing and extraction equipment of nucleic acids from human, animal, and vector respiratory and blood/serum or other biological samples as needed, (3) hybridization-enrichment and amplicon-based MinION and/or Illumina sequencing equipment and supplies, and (4) bioinformatics software and hardware. The hybridization-enrichment sequencing methodology shall contain pathogen-specific enrichment probes customized for the pathogens known or suspected to be endemic at each of the study sites, as determined in collaboration with the NAMRU-PI and local laboratory partners.

5.1.13.3 Conduct molecular sequencing of 50 samples at each laboratory site for the characterization of SARS-CoV-2, Influenza A/B, Respiratory Syncytial Virus (RSV), Dengue Virus, Plasmodium species, Adenovirus, novel human Coronaviruses, other febrile disease, antibiotic resistant or biothreat pathogens as needed, developing quality-controlled genomes of pathogens and vector speciation and identifying variants and sequence polymorphisms in the pathogens and their vectors. Sample testing selection shall be agreed upon with NAMRU 2 and partners/collaborators.

5.1.13.4 Data records shall be maintained and shared only among collaborating institutions and must be approved for presentation or publication by the NAMRU-2 PI in line with NAMRU-2 Public Affairs Officer (PAO) requirements, which may include NAMRU-2 PAO internal approvals. All electronic records shall be stored in a secure computerized system that contains layered passwords for protection and hard copy documents in a locked, secured room.

5.1.13.5 Prepare first draft and assist in the completion of manuscript for research publications based on data collected during the period of performance.

6. SPECIFIC REQUIREMENTS

6.1 NAMRU-2 shall:

6.1.1 Provide access to all documentation associated with the study protocol and related to compliance with NAMRU-2 and local partners human research protection regulations prior to execution of study activities.

7. PLACE OF PERFORMANCE

The work to be performed under this contract shall occur in the USA and countries in the Indo- Pacific region. These countries may include Papua New Guinea, Laos, Malaysia, and/or Vietnam.

8. TYPE OF CONTRACT CONTEMPLATED

The government is considering a non-personal services fully firm-fixed-price (FFP) contract for research and development. The Government shall not exercise any supervision or control over the contract service providers performing the services herein. Such contract service providers shall be accountable solely to the Contractor who, in turn is responsible to the Government.

9. PERIOD OF PERFORMANCE

The period of performance for this contract shall be from SEE SCHEDULE.

10.1 Recognized Holidays

12.2.3 The contractor shall arrange and provide progress updates on all aspects of the contractors’ progress towards completing the work of this contract in a teleconference format on at least a monthly basis to the NAMRU-2 PI, Project Officer, or technical POC. The contractor will generate a brief summary (e.g.

meeting minutes) of the information shared at each teleconference and will provide this in electronic format to NAMRU-2.

12.2.4 Summary reports by request: The contractor shall provide summary reports within 7 days when requested by the PI to assist in providing data to the funding sponsor.

12.3 The contractor shall submit to the NAMRU-2 PI or NAMRU-2 Science Director (SD) for approval prior to public disclosure in line with NAMRU-2 PAO requirements any written publications or public relations materials (including proposed public announcements, flyers, posters, audio-visual materials, newspaper articles, or advertisements) related to this contract.

12.4 The contractor shall assist NAMRU-2 as needed in the preparation of presentations, posters, abstracts, and other materials.

13. RESEARCH CONDUCT

13.1 Research provided under this contract must be conducted in accordance with all state, federal, Department of Defense, Department of the Navy, and command laws, regulations, policies, and procedures that govern the conduct of regulated research. Bureau of Medicine and Surgery (BUMED) Instruction 6500.3 (Series) “Research Integrity, Responsible Conduct of Research Education, and Research Misconduct” applies to research sponsored or funded by Navy Medicine.

Federal regulations governing current Good Clinical Practice (cGCP) are found in 21CFR 11, 50, 54, 56, 312, and 314 and important guidelines are in the FDA Information Sheets. Additional regulations for human subject protection are found in 45CFR 46, and research involving the transportation, care and use of animals should conform to the Animal Welfare Act (7 U.S.C. 2131 et seq.) The International Convention on Harmonization Guidelines for Good Clinical Practice are also standards governing the conduct of research trials. 14. TRAVEL

The Contractor shall complete two trips to train personnel from at least one laboratory site in the Indo-Pacific Region.

15. DATA RIGHTS

15.1 The Contractor shall not release to the public any information regarding the medical research or resulting data without the approval of the Government Surveillance Officer (GSO). The approval process may include blanket approval for some types of information releases.

15.2 The Contractor shall submit to the GSO, PI and Contracting Officer for review and approval prior to public disclosure any written publications or public relations materials (including proposed public announcements, flyers, posters, audiovisual materials, newspaper, advertisements, or mailed advertisements) which are targeted primarily to the scope of contract services or policies and programs of the Contractor.

15.3 The Government has unlimited rights to all documents/material produced under this contract. All documents and materials, to include the source codes of any software, produced under this contract shall be Government owned and are the property of the Government with all rights and privileges of ownership/copyright belonging exclusively to the Government. These documents and materials may not be used or sold by the contractor without written permission from the Contracting Officer. All materials supplied to the Government shall be the sole property of the Government and may not be used for any other purpose. This right does not abrogate any other

Government rights

15.4 As proprietary owner and inventor of this pan-viral panel being tested and evaluated, Dr. Michael Wiley of PraesensBio will likewise maintain rights to this data in order to be used for a future FDA pre-EUA patent application.

16. INVOICING AND PAYMENTS

16.1 All payments will be in US dollars, the United States currency.

16.2 Invoices shall be submitted monthly based on regular report submission.
