About this file
This is a Statement of Need (SON) for the National Institutes of Health to procure a high-resolution laser scanning confocal microscope system to replace aging imaging infrastructure and support biomedical research in hearing and balance.
The Government seeks a research-grade inverted laser scanning confocal microscope system capable of live-cell and fixed-tissue fluorescence imaging with integrated super-resolution imaging, spectral detection, environmental control, automated image acquisition, and quantitative image analysis. The microscope platform must be motorized with high-precision positioning, brightfield and fluorescence imaging capabilities, and a side-port configuration optimized for confocal imaging. The confocal imaging system shall provide spectral laser scanning, detector-array-based super-resolution imaging, high-sensitivity GaAsP detectors, simultaneous multichannel fluorescence acquisition, spectral unmixing, and transmitted-light detection. Required excitation wavelengths include 405 nm, 488 nm, 561 nm, and 639 nm. The system must include research-grade objectives (10×, 20×, 40× oil immersion, and 63× oil immersion), motorized XY stage with high positional accuracy, piezo-driven Z-focusing for rapid volumetric acquisition, and environmental control for long-term live imaging with heated specimen chamber and CO₂ compatibility. The acquisition and analysis software must support automated image acquisition, time-lapse imaging, Z-stack acquisition, tile scanning and stitching, spectral imaging and unmixing, super-resolution image processing, and two-dimensional and three-dimensional image analysis. A high-performance computer workstation with multi-core processor, professional graphics processing unit, ECC memory, RAID storage, current Microsoft Windows operating system, and 4K display is required. The system shall include an air-damped vibration isolation table, all required controllers and cabling, and complete installation and operational verification. The procurement is structured as a Firm Fixed-Price Purchase Order with delivery and installation at NIH's Bethesda, Maryland campus, including assembly, optical alignment, calibration, software installation, system verification, operational testing, and removal of all packing materials. The vendor must provide on-site operational training covering system operation, image acquisition, super-resolution imaging workflows, software operation, routine maintenance, and image processing. Payment shall be made Net 30 following successful delivery, installation, operational testing, and Government acceptance.

STATEMENT OF NEED (SON)
1.0 TITLE
Purchase of a High-Resolution Laser Scanning Confocal Microscope System to Replace End-of- Life Imaging Infrastructure and Support Advanced Live and Super-Resolution Microscopy
2.0 BACKGROUND
The National Institutes of Health (NIH) requires the purchase of a high-resolution laser scanning confocal microscope to replace an aging confocal imaging system that has reached the end of its service life and to support ongoing mission-critical biomedical research in hearing and balance.
The replacement system will support research focused on the cellular and molecular mechanisms underlying hearing and balance using mouse and zebrafish model systems. Current research programs include:
• Live imaging of sensory hair-cell development, regeneration, and function in the zebrafish lateral line and inner ear.
• High-resolution imaging of mouse cochlear and vestibular tissues.
• Visualization of intracellular organelles and protein localization in living and fixed tissues.
• Time-lapse imaging of dynamic cellular processes.
• Quantitative three-dimensional fluorescence microscopy and image analysis.
The required microscope must provide high-resolution spectral confocal imaging with integrated detector-array-based super-resolution imaging, exceptional detector sensitivity, environmental control for live imaging, automated multidimensional image acquisition, and advanced quantitative image analysis. The system must be capable of simultaneously improving spatial resolution and signal-to-noise while permitting imaging at reduced excitation power. These capabilities are essential for long-term live imaging of zebrafish sensory hair cells and mouse inner ear tissues, where minimizing photobleaching and phototoxicity is necessary to preserve specimen viability and obtain quantitative measurements of dynamic cellular processes.
Conventional confocal microscopy cannot provide the combination of spatial resolution, sensitivity, and gentle imaging required for many of these experiments.
The requested system will also provide substantial improvements in imaging throughput and data management. The newer platform produces significantly smaller raw image datasets while dramatically accelerating super-resolution image reconstruction, enabling more efficient acquisition, processing, storage, and analysis of large multidimensional imaging experiments.
