SINGLE SOURCE DETERMINATION USING

SIMPLIFIED ACQUISITION PROCEDURES (SAP)

FOR AN ACTION NOT EXCEEDING THE SIMPLIFIED ACQUISITION THRESHOLD

(AUTHORITY: FAR 13.106-1(b)(1))

1. Agency and contracting activity. Department of Commerce, NOAA Acquisition and Grants Office

2. Description of supplies or services required to meet agency needs (including the estimated value).

1. We require mRNA isolation, library preparation, and Next Generation Sequencing of mRNA (RNA-Seq) isolated from 120 salmon and sablefish tissue samples using the Illumina platforms, i.e. HiSeq or NextSeq, with the vendor providing sample quality scores and final sequences, as fastq files. These products will be used to determine which genes are up or down-regulated in salmon and sablefish exposed to elevated CO2 levels associated with ocean acidification.

Results will be compared to similar gene expression changes in other marine organism exposed to elevated CO2 levels. The vendor must be able to process tissue samples as small as 2 mg. RNA integrity should be assessed by Agilent Tapestation and concentration by Qubit assay. Library size should be assessed on the Agilent Tapestation, and concentration by Qubit assay, and final quantitation by qPCR. Sequencing of the prepared libraries should utilize the Illumina MiSeq, HiSeq or NovaSeq platforms. Sequencing products must be at least ≥80% of bases with ≥Q30. RNA samples must be stored for up to 3 months and FASTQ data stored for up to 6 months following project completion. We anticipate that products will be delivered within 3 months of the contract being awarded.

4. Supporting rationale. Only one source or brand name is reasonably available as detailed below:

□ Compatibility to existing systems or equipment - The required supplies must be compatible in all aspects (form, fit, and function) with existing systems or equipment, and the source is uniquely qualified to meet the requirement.

Azenta Life Science has provided similar RNAseq data for previous NWFSC studies of ocean acidification (OA) effects on marine organisms. Comparisons of how OA affects physiological systems through changes in gene expression require using consistent

RNAseq technologies and methods to make valid comparison between marine organisms.

The specific technologies and lab protocols are unique to Azenta Life Sciences.

5. Market Research.

As indicated, consistency between analytical procedures, materials and platforms is required for comparisons with previous data generated by Azenta Life Science on the effects of OA on gene expression in marine organisms. Our Center studies the effects of ocean acidification on different marine species and in particular, we are trying to understand the physiological processes that underly sensitivity or resilience to future ocean acidification (OA) conditions. It is important to understand how different organisms may use similar or different mechanisms to deal with these conditions. Our colleagues at the Center conducted a very similar study exposing Littleneck clams (a marine invertebrate model) to elevated CO2 levels (driver of OA) and then examined the impacts on survival and ability to thrive, while in parallel, examined which physiological processes are altered (using the RNAseq techniques described in our purchase request). We are following up on these studies, to examine these similar physiological pathways in marine vertebrate species (salmon and sablefish) using the same techniques.

While many companies and vendors offer quite similar services, each have unique laboratories, equipment, protocols, and materials. In order to make valid comparisons between our two experiments, it is critical that we employ the same vendor (i.e., methods, materials, etc.) for these studies. Future experiments that don’t involve this direct, between-experiment assessment may not require the same vendor.
