About this file
This justification document requests approval for a sole-source acquisition of 10X Genomics Chromium Single Cell reagents from 10X Genomics. The National Institute of Allergy and Infectious Diseases (NIAID) within the National Institutes of Health (NIH) requires the reagents to conduct single cell RNAseq experiments to evaluate changes in cell populations at the transcriptome level. The reagents allow separation and labeling of single cells so that gene expression profiles can be connected to individual cells. This exclusive technology is only available from 10X Genomics and is needed to maintain continuity of data across experiments. If approved, NIAID will acquire $26,385.50 worth of reagents from 10X Genomics on a non-competitive basis using the statutory authority that only one responsible source is available.

Justification for Other than Full and Open Competition at or Below the Simplified Acquisition Threshold Project Title: 10X Genomics Chromium™ Single Cell Reagents
I . Identification of the agency and contracting activity
DHHS, NIH, NIAID, DIR, LHIM
Requestors name:
Requestors phone number:
Requestors email address:
Address of the requesting lab/office: NIH/NIAID/Metaorganism Immunity Section, 4 Memorial Drive, Bethesda, MD, USA 20892.
II. Description of the project, and supplies or services required to meet the agency’s needs (including the estimated value)
A. Type of action. Check the appropriate box:
☒ New requirement ☐ Follow-on
• Name of the previous contractor(s): __10X Genomics__________________
• Sequence number(s), if available: _______________
• Was the initial and any subsequent awards competed? ☐ Yes ☒ No
B. AMBIS Sequence number for this requirement: _2177598___ C. Total estimated dollar value of the requirement (including options, if applicable): _ $ 26,385.50 D. Period of performance, inclusive of options: from ___________ to ________________ E. Project Background
LHIM scientists broadly study the factors controlling immunity and inflammation, and utilize transdisciplinary approaches in the exploration of host-microbe, tissue-tissue, and host-environment interactions. This includes microorganisms such as Leishmania major, Toxoplasma gondii, and Cryptosporidium species. A major goal is to assess the consequences of environmental stress and inflammatory disorders, as well as explore novel microbiota-mediated and immunotherapeutic approaches to combat the increasing threat of microbial resistance. This work employs investigations in cell culture, animal models, immune responses, and cellular and molecular biology.
F. Project Description LHIM uses cell culture, animal models of disease, and cellular and molecular biology techniques. The use of RNAseq technologies are a powerful tool to understand the changes in gene expression over time, but currently, this is done in one reaction so that you can’t ascribe the signaling to one cell or another. Many of LHIM’s projects require single cell RNAseq experiments to evaluate changes in the transcriptome of diverse cell populations. The 10X Genomics Chromium Single Cell reagents allow you to separate and label single cells so that now the gene expression profiles can be connected to one cell. This is powerful new technology that then gives you information as to the identity of the cell, whether it is directly infected, and what its gene expression profiles are. This exclusive technology requires reagents available solely through 10X Genomics. It is also needed to keep continuity of data across experiments.
III. Identification of the statutory authority permitting other than full and open competition (SELECT ONE) ☒ 41 USC 253(c)(1) a n d F A R P a r t 6 . 3 0 2 - 1 : Only one responsible source ☐ 41 USC 501 a n d F A R P a r t 8 . 4 0 5 - 6 : Federal Supply Schedule (FSS), limited competition
☐ An urgent and compelling need exists, and following the procedures would result in unacceptable delays
☒ Only one source is capable of providing the supplies or services required at the level of quality required because the supplies or services are unique or highly specialized; or
☐ In the interest of economy and efficiency, the new work is a logical follow-on to an original Federal Supply Schedule order provided that the original order was placed in accordance with the applicable Federal Supply Schedule ordering procedures. The original order or BPA must not have been previously issued under sole-source or limited-sources procedures.
Important Note: The statutory authorities listed below will rarely ever be used in NIH. Before selecting one of these authorities, please ensure that you have read and understood the citation and discussed it with the cognizant Contracting Officer.

IV. Demonstration that the proposed contractor(s) unique qualifications or the nature of the acquisition requires use of the authority cited.
A. Name and address of the proposed contractor(s): 10x Genomics, 6230 Stoneridge Mall Road, Pleasanton, California, USA 94588-3260.
B. Describe why only the requested supply/service will meet the government’s needs.
Many of LHIM’s projects require single cell RNAseq experiments to evaluate changes in the transcriptome of diverse cell populations. The 10X Genomics Chromium Single Cell reagents allow you to separate and label single cells so that now the gene expression profiles can be connected to one cell. This exclusive technology is only available through 10X Genomics. It’s also critical that LHIM continue to use 10X Genomics Chromium Single Cell reagents to keep continuity of data across experiments.
C. Describe why only the proposed contractor is capable of providing the requested supply/service.
10x Genomics is the only manufacturer and distributor of the Chromium Single Cell reagents.
V . Determination by the Contracting Officer that the anticipated cost/price to the Government will be fair and reasonable based on
VI. Certification This is to certify that the justification for the proposed acquisition has been reviewed and that to the best of my knowledge and belief the information and/or data provided to support my request (for the requestor) and approval (for the CO) of is accurate and complete.
