About this file

This is a Request for Information (RFI) issued by the Defense Health Agency (DHA) seeking industry input on the development and regulatory pathway for Dimethyl Sulfoxide (DMSO) Cryopreserved Platelets (CPP), a frozen biologic platelet product intended for US FDA licensure. The RFI was issued on 23 April 2026 and is managed by DHA's Operational Medical Systems, Warfighter Protection and Acute Care Project Management Office (WPAC PMO) at Fort Detrick, Maryland, with submissions directed to Peter Mitchell at the Defense Health Agency Contracting Agency.

The technical requirement centers on sponsoring and executing all activities necessary to serve as the Investigational New Drug (IND) application holder and regulatory sponsor for IND #14047, with performance of all FDA recommendations and procedures applicable to Emergency Use Authorization (EUA) authorization, culminating in submission of a Biological License Application. DMSO CPP offers a shelf life of two years or potentially longer compared to conventional liquid stored platelets limited to 5-7 days at room temperature. Previous Phase 1 and Phase 2 clinical studies have demonstrated in vivo viability and non-inferiority to liquid stored platelets in treating postoperative blood loss. The estimated period of performance for the EUA phase is no longer than 60 months. Responses must include a white paper not exceeding 10 pages, a rough order of magnitude cost estimate, and a completed contact information table demonstrating the responder's technical expertise, FDA experience, and capability to perform regulatory sponsorship activities. All responses must be submitted electronically in Microsoft Word or searchable Adobe PDF format to peter.d.mitchell8.civ@health.mil with subject line "Response to RFI for Cryopreserved Platelets (CPP)" no later than 3:00 PM Eastern Time on 4 May 2026. The RFI explicitly disclaims any obligation to award a contract or conduct a future solicitation, and the Government reserves the right to use all submitted information for any legally appropriate purpose.

7.0 DESCRIPTION: The mission of DHA OPMED is to develop and manage medical materiel to protect and sustain the Warfighter on point for the Nation. We develop, directly through collaboration with commercial partners, medical products that enhance readiness and improve the quality of life of active-duty personnel in the operational environment. OPMED’s WPAC PMO has been developing a biologic product: Dimethyl Sulfoxide (DMSO) Cryopreserved Platelets (CPP), a frozen, biologic-platelet product. DMSO CPP are formulated to provide a platelet-derived product with a longer shelf life (2 years and potentially longer) than liquid stored platelets (LSP), which are currently licensed for storage up to 5-7 days at room temperature (20ºC to 24ºC) under gentle agitation.

In vitro studies demonstrate that compared to LSP, CPP is potentially more hemostatically active. Such characteristic is defined by irreversible changes to platelets activation, microparticle release, loss of in vitro functions including a reduced response to agonists and hypotonic stress, decrease in shape change ability, increase in thrombin generation, and a conversion to a procoagulant phenotype (PS expression) and increased numbers of microparticles. DMSO CPP are prepared from γ-irradiated [2500 centi-gray (cGy)] leukocyte-reduced apheresis platelets, stored in plasma and anticoagulant citrate dextrose solution A for up to 57 hours at room temperature (20°C to 24°C) after apheresis. The apheresis platelets are then concentrated and cryopreserved in approximately 6% DMSO at -80°C and stored at ≤-65°C.

OPMED’s past and current industry and academic partners have manufactured clinical-grade lots of CPP and performed Phase 1 and Phase 2 clinical studies under a US Investigational New Drug (IND) application. Several Phase 1 in vivo recovery and survival studies demonstrated in vivo viability of CPP product as required by FDA’s standards. In a Phase 1/2a dose-escalation study, no thromboembolic complications resulted from the transfusion of up to 3 units of CPP into actively bleeding thrombocytopenic patients. In a Phase 2, in adult patients undergoing Cardiopulmonary Bypass Surgery, CPP was noninferior to LSP in treating postoperative blood loss.

Emergency Use Authorizations (EUAs) are vital to development by accelerating access to critical medical products during public and military health emergencies, ensuring that potentially life-saving tools are available faster than standard approval processes allow. EUAs balance rapid deployment with safety, relying on robust, albeit preliminary, evidence to manage risks during crises.

7.1 Technical Aims

7.1 Performance of all FDA’s general recommendations and procedures applicable to the authorization of an EUA.

7.2 Sponsorship and execution of all activities commensurate with serving as the Investigational New Drug (IND) application # 14047 holder and regulatory sponsor in accordance with 21 Code of Federal Regulations 312 subpart D, including all regulatory submissions and continuation through submission of the Biological License Application.

The EUA period of performance is estimated to take no longer than sixty (60) months.
