Statement of Work

Program Office:National Center for Advancing Translational Sciences, NCATS
National Institutes of Health
9800 Medical Center Drive, Building B, Room B3005
Rockville, MD 20850

Project Title: Acquisition of testing services: twenty (20) compounds assayed in a biochemical assay panel and thirty (30) compounds vs KDM4A, KDM5A, KDM5B, KDM6B, and KMD1a Background The National Institutes of Health (NIH) is the nation’s leading medical research agency and the primary Federal agency whose mission is to seek fundamental knowledge about the nature and behavior of living systems and the application of that knowledge to enhance health, lengthen life, and reduce illness and disability, conducting, supporting and making medical discoveries that improve people’s health and save lives.

The National Center for Advancing Translational Sciences (NCATS) is a part of the National Institutes of Health (NIH), which mission it is to catalyze the generation of innovative methods and technologies that will enhance the development, testing and implementation of diagnostics and therapeutics across a wide range of human diseases and conditions.

The NCATS Chemical Genomics Center (NCGC) is tasked with identifying and developing novel small molecule leads against a wide spectrum of human disease targets, which it accomplishes by screening thousands of small molecules in collaboration with both intra-and extramural researchers. In pursuit of this goal, the Center has a strong internal transdisciplinary team of scientists who continually discover novel small molecules for application against a wide range of human diseases.

As such, discovering probe molecules that could serve as starting points for therapeutics in rare cancers is of great interest within NCATS. NCATS has several projects that address many aspects of oncology mechanisms and approaches to discovering novel, effective small molecules. The services requested here are fee for testing services that will provide insight on efficacy of compounds across many cancers and will help progress and provide critical scientific guidance on projects that NCATS has been working on for many years. Reaction Biology is the only vendor who offers a panel consisting of 78 kinases, 11 phosphatases, 63 proteases, 35 MTs, 5 demethylases, 28 reader domains in AlphaScreen, 88 reader domains in Thermal Shift, 11 HDACs, 4 SIRTs, 7 HATs, 12 DUBs, 3 PARPs, 12 PDEs, 2 Bcls, 5 CYPs, herg, 2 HSP90s, 6 IDHs, and MTH and hence we are including a sole source justification as part of the package. Futhermore, we are in need of IC50 determinations for 30 compounds against KDM4A, KDM5A, KDM5B, KDM6B, and KDM1a.

Statement of Objective This request is for fee for testing services to provide the required knowledge of each agent’s polypharmacology. The data obtained from these assays will be used to establish an understanding of the contributing pharmacology to specific stem-cell differentiation events.

Scope of Work Vendor will need to provide the following services:

· Assay 20 compounds against biochemical assay panel that includes 578 kinases, 11 phosphatases, 63 proteases, 35 MTs, 5 demethylases, 28 reader domains in AlphaScreen, 88 reader domains in Thermal Shift, 11 HDACs, 4 SIRTs, 7 HATs, 12 DUBs, 3 PARPs, 12 PDEs, 2 Bcls, 5 CYPs, herg, 2 HSP90s, 6 IDHs and MTH

· Vendor shall perform each assay with a single dose in duplicate

· The specific assays requested are outlined in the attached assay document

· Assay 30 separate compounds to determine IC50 against KDM4A, KDM5A, KDM5B, KDM6B, and KMD1a

· IC50 is to be obtained using 10 doses in a singlet assay

· Vendor shall provide detailed assay results with four (4) weeks upon receipt of material from NCATS.

Deliverables / Reporting Requirements:

The contractor is required to provide written report on all data generated within 4 weeks to project officer (TBD), 9800 Medical Center Drive, Rockville, MD 20850.

Period of Performance These studies should take no longer than 4 weeks upon receipt of materials from NCATS and services are considered complete once shared through the secure web-enabled sharing resource.

Section 508 Electronic and Information Technology Standards N/A

CONTRACTING OFFICER'S REPRESENTATIVE (COR)

The following Contracting Officer's Representative (COR) will represent the Government for the purpose of this contract:

TBD

Mandatory Criteria (Requirements) The contractor will be evaluated on the following:

· Technical Approach (75%)

· The contractor shall demonstrate their understanding of the requirements and tasks detailed in the scope of work and their technical approach to perform the required tasks outlined.

· Price Competitiveness (25%
