Questions received for: Generation and characterization of modified peptides and generation of monoclonal antibodies specifically recognizing these modified peptides
Q: NIA must provide protected, modified peptides to a company with experience in the specific deprotection procedures that are required to not damage our modified peptides. What are the peptide sequences? What type of deprotection method is required?
A: Several deprotection methods are required including reduction of disulfide bonds, as well as more complex procedures to protect modified amino acids that are not very abundant. Because of challenges with abundance of our modified peptides these deprotection reactions may need to be monitored by LC-MS and possibly HPLC purified after deprotection
The peptide sequences are confidential.
Q: By default, we will clone single clones (mAb expressing hybridoma cell lines). If purified antibodies are needed as well, what quantity, and how many antibodies?
A: Clone supernatants tested for selectivity by ELISA are required for this quote.
