BACKGROUND

When flash chromatography is currently used, a glass cylinder is manually filled with stationary phase (e.g., silica gel), equilibrated with a solvent system, the sample solution is loaded on top of the stationary phase, and elution is performed manually by slight air compression of the eluent. The process is time-consuming and has the additional drawback that during the separation and collection of fractions, vapors of solvents may be inhaled by the chromatographer. An automatized flash chromatography system is needed to significantly shorten the process of separation and improve the safety of the operation.

REQUIREMENTS

• Two column channel flash purification system for instantaneous switching between normal and reverse phase column purification

• Smallest in class dimensions: 13.2” x 15.5” x 21.5” (Width x Depth x Height)

• 15” solvent stable TFT LCD Touch Screen

• 4 x 4L solvent containers capable to fit on top of Selekt system using solvent containment system accessory (included in this quote)

• Ethernet port, 4 USB ports, 2 D-Sub 9-pin ports, and AUX port

• Real-time PDA display of eluting compounds for instant purity monitoring and assessment

• 2D and 3D spectra analysis displays for purity assessment

• λ-All feature improves sensitivity and detects any light-absorbing compound reducing sample loss

• Baseline correction capability eliminates baseline rise/fall during gradients

• TLC-to-linear and TLC-to-step gradient method capability with cartridge and sample load recommendations

• Gradient optimization (GO function) isolates the compound of interest in 50% less time and solvent

• U/V Variable (198-810nm). Amplitude range: 0-6400 mAU (Absorbace Units);

accuracy of +/- 5 mAU.

• No sample loss when changing fractions

• Purify mg to 75+ grams on a single system without method re-optimization

• 1-300 mL/min flow rate with accompanying Sfär columns, in 1 mL/min increments

• Pressure range of 0-30 bar (0-3000 kPa: 0-435 psi)

• Fractionate using two different wavelengths at the same time (UV1 & UV2) & monitor/collect on λ-All

• Software compatibility with Biotage Sfär, Snap Ultra, and ZIP Flash

Chromatography Cartridges

• β built-in system flushes with option to collect the effluent to prevent sample loss and cross contamination.

• Gradient precision of +/- 2%

• External fluid connections to simplify visual leak detection and repairs

Packaging and Marking

• The instrument shall be delivered with all necessary supplies and accessories required for the installation and start-up. Cost of installation and user-familiarization training shall be included in the price.

• The instrument shall be covered under warranty for one year after installation.

DELIVERY LOCATION

The system that meets above requirements or its equivalent and other accessories shall be complete and shall be delivered to:

U.S. Food and Drug Administration Office of Cosmetics and Colors, Room 1069 4300 River Road College Park, MD 20740

Schedule: Delivery shall be no later than 30 days after contract award

INSPECTION AND ACCEPTANCE CRITERIA

The Government shall have 30 working days to inspect and verify that the equipment and its operational software are functional and demonstrate performance.

PERIOD OF PERFORMANCE

The period of performance is 12 months from the successful completion of installation of software, hardware, and training of FDA scientists. This order also includes four (4) one

(1) year option periods.

CONTRACT ADMINISTRATION

The Contracting Officer’s Representative (COR) will perform inspection and acceptance of equipment, installation and services to be provided.

The COR is responsible for the following as required by this order: (1) monitoring the Contractor's technical progress, including the surveillance and assessment of performance and recommending to the Contracting Officer changes in requirements; (2) interpreting the Statement of Work and any other technical performance requirements; (3) performing technical evaluations; (4) performing technical inspections and acceptances; and (5) assisting in the resolution of technical problems encountered during performance.
