Human Form of CaV1.2 Channels for In Vitro Electrophysiology/Ion Channel Pharmacology Project
Notice Type: Combined Synopsis/Solicitation
This is a combined synopsis/solicitation for cultured cells stably expressing human form of CaV1.2 channels for in vitro electrophysiology/ion channel pharmacology project. This is for commercial items prepared in accordance with the format in Federal Acquisition Regulation (FAR) Subpart 12.6 “Streamlined Procedures for Evaluation and Solicitation for Commercial Items”, as supplemented with additional information included on this notice and in conjunction with FAR Part 13.106, utilizing Simplified Acquisition Procedures. This announcement constitutes the only solicitation; quotes are being requested and a written solicitation will not be issued.
Background: The U.S. Food and Drug Administration (FDA), Division of Applied Regulatory Science (DARS) has established a research program for evaluating the safety and efficacy on drug pertaining to cardiac functions, and the experiments outline in the research program involve recording how various drugs affect the activities of cardiac ion channels expressed in heterologous systems (such as CHO and HEK293 cells). The CaV1.2 channels (L-type Ca2+ channels) are a major type of cardiac ion channels that contribute to shape the cardiac action potentials and initiate cardiac muscle contraction. Hence, activities of these channels are tightly linked with cardiac output. Given that drugs that act on CaV1.2 channels can alter cardiac functions, for DARS’ ongoing cardiac safety research program. FDA, DARS require the purchase of a cell line that stably expresses the CaV1.2 channel proteins for the ion channel pharmacology experiments.

Technical Requirements:
The vendor shall provide one or more vials of cryopreserved, division-competent cells that stably express the human versions of CaV1.2 protein (alpha or pore-forming subunit), beta 2 protein (auxiliary subunit), and alpha-2-delta protein (also auxiliary subunit). Note that the expression of the auxiliary subunits (not just the alpha subunit) in these cells is critical, because 1) the expression of alpha and auxiliary subunits reflect in vivo configuration of the CaV1.2 channels; 2) auxiliary subunits regulate CaV1.2 channel activity; and 3) drugs that affect cardiac functions by acting on CaV1.2 channels can do so by binding to channel alpha and auxiliary subunits.
