SECTION III: ADDENDA TO FAR CLAUSE 52.212-4, CONTRACT TERMS AND

CONDITIONS –COMMERCIAL ITEMS (FEB 2012)

III.1. PERFORMANCE SPECIFICATIONS/REQUIREMENTS

III.1.1 Minimum Performance Specifications- Confocal/Multi-photon Microscope

The Food and Drug Administration’s (FDA) Confocal Microscopy Core Facility of the Center for Biological Evaluation and Research (CBER) supports all FDA scientists and laboratories for their morphological studies and light microscopic imaging. The facility’s primary purpose is to analyze host – pathogen (virus, bacteria) interaction using cultured cells, tissue and small animals in immunological response and vaccine related studies. The Core Facility requires a new point scanner Confocal/Multi-photon (MP), super resolution microscope system that has the following modules/applications to accommodate CBER investigators’ broad imaging needs which are critical for the facility’s research/review programs:

1. New microscope with a high resolution, high sensitivity and high speed live confocal scanning features, with efficient multicolor image acquisition from live or fixed cell or tissue samples.

2. Shall be capable of molecular mobility assay such as fluorescence recovery after photo-bleaching (FRAP), fluorescence correlation spectroscopy (FCS). The system must have ability to track/visualize both very fast and slow movement of fluorescently labeled peptide or viral particles and flexible/variable scan speed depend on imaging projects.

3. Shall be able to analyze single molecule mobility and molecular interaction measurement in Fluorescent Protein expressed living cells for in situ biochemistry assay.

4. Fully tunable true white light laser (WLL) with up to active 8 simultaneous lines.

5. Flexible excitation wavelength between 470-670 nm for conventional confocal microscope applications. The system shall have the ability to acquire excitation spectra/curve from any unknown dye, fluorescence proteins or fluorochrome to select the best excitation for FRAP, FRET, FCS and multicolor experiment.

6. Molecular interaction assay modules such as acceptor bleaching, sensitized emission, spectral and fluorescent lifetime (FLIM), and Förster resonance energy transfer (FRET).

7. Lamda spectral scan capability for detecting the full emission wavelength spectra/curve for dye separation and ready for multicolor analysis for handling new dye, fluorochrome or fluorescence proteins.

8. Capability for at least 4 channels intra-vital multi-photon and second harmonic generation (SHG) microscopy. It shall be equipped with two external non-descanned detectors (NDD) for sensitive imaging projects for flexible multicolor intra-vital microscopy and ready for Coherent Anti- Stokes Raman Scattering (CARS) and uncaging microscopy.

9. Super resolution Microscopy (nanoscopy) with the ability to identify very tiny objects (about 50 nm). The system shall be able to visualize live RNA and/or DNA viruses, bacteria, cell receptors or nano particles. This system shall have the ability to perform at least two channel super resolution microscopy (nanoscopy) with 50 nm resolution that has capability for visualizing budding of influenza A virus, Hepatitis A, West-Nile, mumps and other viruses from live cell without post image processing. Simultaneously, the system shall be able to perform confocal scanning in 3 other channels.

10. Shall be an inverted microscope, ready for applications of live cells or animals equipped with large environmental chamber with CO2 and temperature control modules(4-37 °C).

11. High content microscopy capability, ready for fully automated autofocus, large tile scan, or 5D multi point time lapse imaging on many vaccine and drug candidate screening applications with real time data streaming/control.

12. 2-3 days of on-site training at Government facility for 3-5 users (training shall cover entire system- microscope + laser).

13. Purchase of the system shall include a minimum of 12 months of upgrades/updates and a minimum of 12 months of phone and email support from the date of acceptance.

14. Vendor must provide OEM-certified technicians for maintenance service and support for the base year and option years of the contract.

15. Automated staining accessories for multiphoton immunofluorescence sample preparation.

III.1.2 Minimum Performance Specifications for Infrared-Optical Parametric Oscillator (IR-OPO) Laser:

The CBER Confocal Microscopy Core facility requires an IR-OPO laser system that is compatible with the aforementioned confocal microscope. The IR-OPO laser system shall accommodate CBER investigators’ intra-vital multiphoton (MP), second harmonic generation (SHG), Coherent anti-Stokes Raman spectroscopy (CARS) imaging needs. When added to the confocal microscope, the combined system will be used primarily to analyze the host – pathogen (virus, bacteria) interaction using intravital MP, SHG CARS technologies on small animals, imaging for viewing of immunological response and vaccine related studies. Minimum performance characteristics for the laser component are as follows:

1. Combination of ultrafast IR-OPO laser and ultrafast IR laser system for 4 color multiphoton, second harmonic generation (SHG) and Coherent anti-Stokes Raman spectroscopy (CARS) microscopy.

2. Optical parametric oscillator (OPO) which can produce 100fs ultra-short laser pulse with high peak power and tunable between 680-1300 nm. It shall deliver the femto-second laser pulse to the deep tissue for multi-photon, SHG and CARS imaging, e.g. animal lymph nodes, spleen, lung, intestine, muscle and brain. It shall be ready for applications of multi-photon microscopy, femtosecond Coherent anti-Stokes Raman spectroscopy (CARS) and uncaging experiment.

3. Ability to produce sub70fs ultra-short laser pulse with high peak power and tunable between wavelength 690 nm to 1040 nm with high power for deep tissue multi-photon and SHG imaging, e.g. animal lymph nodes, spleen, lung, intestine, muscle and brain.

4. Simple beam routing system for daily maintenance and easy interface for multi-user setting.

5. Group-velocity dispersion (GVD) compensation system for better deep tissue imaging. This pulse compensation system is critical for ultimate deep tissue multiphoton SHG animal imaging.

6. Active anti-vibration table.

7. Fully compatible with the vendor’s proposed confocal microscope system.

8. Minimum one-year full system warranty which includes all optics, optical coatings, diodes, mechanical, electronic components, chiller and power supply to ensure specified system performance

9. Field serviceable beyond a diagnostics-level so that most repairs and maintenance can be done without removing the laser from the experimental setup and without the requiring the laser to be sent to the laser manufacturer's factory for repair.

Pump laser head shall be removable and field replaceable in order to minimize downtime.

10. Purchase of the system shall include a minimum of 12 months of upgrades/updates and a minium of 12 months of phone and email support from the date of acceptance.

11. Vendor must provide OEM-certified technicians for maintenance service and support for the base year and option years of the contract.
