PART 3 – Description/Specifications

3.1 Background

LDV’s research focuses on developing new methods to measure the potency of seasonal and pre-pandemic influenza vaccines, and the design and development of in-vitro binding and competition assays to evaluate the immune response to SARS-CoV-2 and influenza vaccines.

We are interested in producing strain-specific monoclonal antibodies that are used in conjunction with strain-specific antigen standards in current influenza potency assays. In addition, we are interested in developing monoclonal antibodies that are type-specific so that they can be broadly used to assay the potency of all vaccine strains of a particular influenza subtype.

There is a need for a deeper understanding of how the human body mounts an immune response to the SARS CoV-2 virus. We are interested in developing monoclonal antibodies against SARS-CoV-2 antigens. This includes antibodies against the Spike protein receptor-binding domain (RBD) for binding and competition assays that will use immune sera.

3.2 Objectives

The objective of this requisition is to obtain services for the delivery of isolated Monoclonal hybridoma on a ‘best effort’ basis.

3.3 Scope

Independently and not as an agent of the Government, the Contractor shall furnish the necessary personnel, materials, services, facilities, and otherwise do all things necessary for or incidental to the performance of the work as described below.

3.4 Tasks

3.4.1 Task 1 – Stage 1 Immunization

A minimum of (5) mice will be immunized with an antigen provided by the FDA. Vendor will use an appropriate adjuvant.

3.4.2 Task 2 – Stage 2 Serum Screening

Sera from mice will be screened by the vendor using ELISA to the antigen provided by FDA, mice with the best antibody titers will be boosted and selected for fusion.

3.4.3 Task 3 – Stage 3 Fusion

Spleen cells of the selected mice will be fused with a myeloma cell line to generate hybridoma cells expressing monoclonal antibodies. Six to ten 96-well plates will be seeded with the fusion products.

3.4.4 Task 4 – Stage 4 Fusion Screening

Fusion lines will be screened by the vendor using an ELISA against an FDA-supplied antigen. Because it is not possible to predict the success of a fusion to generate hybridomas with the desired antibody characteristics, two separate fusions, from a minimum of two individual immunized mice may be necessary.

3.4.5 Task 5 – Stage 5 Expansion

Approximately 25-50 positive wells will be selected for expansion, cloning, and further testing. Serum-free supernatants (~ 5-10 mL/clone) will be supplied to FDA for further screening to select the desired clones. Five (5) hybridoma clones selected by FDA will be expanded, cloned, and used for antibody production. A second splenocyte fusion from a second immunized mouse will be performed if necessary.

3.4.6 Task 6 – Stage 6 Antibody Production and Purification

Antibody production will be performed in-vitro (tissue cell culture) to generate ten (10) mg of purified monoclonal antibody. Purification of IgG will be done using Protein A or Protein G affinity purification. Antibody concentration should be no less than one (1) mg/mL. Final endotoxin content of the purified antibody should be less than ten (10) EU/mL. Purified mAb will be delivered to FDA. Hybridoma cells will be tested for mycoplasma and frozen in liquid nitrogen for long-term storage. Frozen hybridoma cells (minimum of five (5) vials/clone) will be delivered to FDA, along with a certificate showing that they are mycoplasma-free.

Part 4 –Performance and Deliverables

4.1 Deliverables

Because it is not possible to guarantee the successful isolation of a monoclonal hybridoma with the desired reactivity, all work will be done on a ‘best effort’ basis.

By the end date of the period of performance, the contractor shall deliver:

1) Five (5) vials of frozen cells for each of the final subclones with positive reactivity to the provided immunizing antigen (five (5) clones as selected by the FDA) will be delivered to the FDA.

2) 10 mg of purified IgG produced in cell culture from each of the final subclones with positive reactivity to the provided immunizing antigen (five (5) clones as selected by the FDA) will be delivered to the FDA.

3) Supernatants will be provided to the FDA for screening during the project as outlined in the technical requirements above.

Delivery Location:

U.S. Food and Drug Administration CBER/OVRR/Division of Viral Products 10903 New Hampshire Avenue Building 72, Room 1366 Silver Spring, MD 20993 Attention: Falko Schmeisser Email: falko.schmeisser@fda.hhs.gov Phone: 240-402-7364

4.2 Work Hours

The contractor shall work hours as necessary to accomplish the contract objectives, generally an 8-hour period between 8:00 am and 6:00 pm. Unless otherwise directed, the contractor shall adhere to the standard work hours, working no more than 40 hours per week and observe all U.S. Government holidays.

4.3 Packing, Marking and Shipping

At a minimum, all deliverables shall be marked with the contract number and contractor name. Deliverables shall be securely packaged and frozen in liquid nitrogen for long-term storage along with a certificate showing they are mycoplasma-free. The contractor shall guarantee that all required materials shall be delivered in acceptable condition.

4.4 Inspection and Acceptance

The Contracting Officer Representative (COR) will perform inspection and acceptance of deliverables and services to be provided. The COR – to be determined upon contract award - is the authorized representative of the Contracting Officer.
