U.S. DEPARTMENT OF COMMERCE

National Oceanic and Atmospheric Administration Eastern Acquisition Division Norfolk Federal Building 200 Granby Street, Room 815 Norfolk, VA 23510

SINGLE SOURCE DETERMINATION USING

SIMPLIFIED ACQUISITION PROCEDURES (SAP)

FOR AN ACTION NOT EXCEEDING THE SIMPLIFIED ACQUISITION THRESHOLD

(AUTHORITY: FAR 13.106-1(b)(1))

NFFM7420-20-02590

1. Agency and contracting activity. Department of Commerce, NOAA Acquisition and Grants

Office (AGO), Eastern Acquisition Division (EAD) and National Marine Fisheries Services NMFS), Northeast Fisheries Science Center (NEFSC) Aquaculture Sustainability Branch (ASB)

2. Description of supplies or services required to meet agency needs (including the estimated value). (Provide a description of the supplies or services required, to include intended use, and the expected delivery date/period of performance. Identify the contract type planned and provide the total estimated value, including all options.)

Provide DNA Sequencing Analysis of cultures from the Milford Laboratory Microalgal Culture Collection and work with the Institute to obtain sequence draft genome sequences for accurate strain identification and the development of strain-specific genetic tests for future collection assessment.

3. Identification of the single source or the brand name to be solicited. (Name, address, POC, phone number.)

University of Connecticut DUNS: 927570622 67 North Eagleville Road Unit 3197 Engineering Science Building 306B Storrs, CT 06269-3197 POC: Dr. Rachel O’Neill Email: rachel.oneill@uconn.edu

PH: 860-486-6031

4. Supporting rationale. Only one source or brand name is reasonably available as detailed below:

(Check all applicable boxes and provide supporting rationale under the selection. Delete choices not selected.)

□ Other - Only one source is capable (Identify any other reason not identified above as to why only the single source is capable of providing the supplies or services required at the level of quality required because the supplies or services are unique or highly specialized. Provide details on mission impact if the requirement is not solicited from the single source.)

Dr. Rachel O’Neill’s laboratory at the University of Connecticut is fully equipped for the necessary steps to prepare the cultures for DNA sequencing. The protocol for the unique sequencing approach was developed in her laboratory. This R&D supports collaboration on applied research that builds upon basic research to exploit the potential of Dr. O’Neill’s scientific discoveries, to advance the state of the art in service to NEFSC aquaculture objectives.

Microalgal cultures selected for DNA sequence analysis will be transported to Rachel O’Neill’s laboratory at UCONN, which is fully equipped for the necessary steps to prepare the material for sequencing. Genomic DNA from 7-10 different sample collections will be extracted using a high molecular weight (HMW) extraction protocol developed in the O’Neill laboratory that is specific for data acquisition of long-read sequences (10-200 KB stretches of DNA, where 1 KB = 1,000 nucleotides).

The O’Neill Laboratory at the University of Connecticut has developed a high molecular weight protocol that is specific for the data acquisition of long-read sequences required for our project.

5. Market Research.

(Provide a brief description of the results of the market research performed. The extent of market research should be commensurate with the total estimated value. If a market research report is available, provide as an attachment.)

No other known source can supply this DNA analysis to our specifications as the O’Neill Laboratory. The long-read technique that will be readable on a portable, hand-held device to confirm strain identity is an emerging capability unique to the O’Neill laboratory.

**Signatures shall remain on the final page.

6. Technical/Requirements Representative Certification

I certify that this requirement constitutes the Government’s minimum needs and the supporting data provided herein is accurate and complete to the best of my knowledge and belief.
