STATEMENT OF WORK

FOR

DEPARTMENT OF THE ARMY

RESEARCH, DEVELOPMENT AND ENGINEERING COMMAND

INTERNATIONAL TECHNOLOGY CENTER-PACIFIC

(ITC-PAC)

Improvement of Cell-based Therapy for Bone Injury (Document No. W90GQZ-71860040)

1. DESCRIPTION OF SERVICE.

The proposal is to develop technologies to improve non-viral methods for direct conversion of fibroblast cells into osteoblasts by transducing genes, and to improve the induction of Directly Converted Osteoblasts (dOBs) under xeno-free conditions leading to improved cell-based therapy for bone injury.

Due to some recent new findings by the PI researcher that retrovirus-mediated transduction of a particular combination of osteoblast-specific transcription factors and reprogramming factors drastically converted approximately 80% of human fibroblasts into osteocalcin-expressing cells that produced mineralized bone matrix. The directly converted osteoblasts (dOBs) displayed similar gene expression profiles as human osteoblasts, and contributed to bone repair after transplantation into NOD/SCID mice at an artificial bone defect lesion of the femur. Therefore, in this new project, he is going to improve the non-viral procedures of conversion, as well as conversion procedure under xeno-free conditions, to establish more efficient and clinically feasible dOBs for bone defect and fracture.

Duration of this Research and Development is for a 12-month period.
