NON-COMPETITIVE

SEQ CHAPTER \h \r 1COMBINED SYNOPSIS / SOLICITATION

Title:

Electrophysiological Characterization of Ion Channels of iPSC Derived Sensory Neurons (i) This is a combined synopsis/solicitation for commercial items prepared in accordance with the format in Subpart 12.6 as supplemented with additional information included in this notice. This announcement constitutes the only solicitation; proposals are being requested and a written solicitation will not be issued.

(ii) The solicitation number is 75N95020Q00267 and the solicitation is issued as a request for quotation (RFQ).

This acquisition is for a commercial item or service and is conducted under the authority of the Federal Acquisition Regulation (FAR) Part 13—Simplified Acquisition Procedures; and FAR Part 12—Acquisition of Commercial Items, and is not expected to exceed the simplified acquisition threshold.

THIS IS A NON-COMPETITIVE (NOTICE OF INTENT) COMBINED SYNOPSIS SOLICITATION TO AWARD A CONTRACT OR PURCHASE ORDER WITHOUT PROVIDING FOR FULL OR OPEN COMPETITION (INCLUDING BRAND-NAME).

The National Institute on Drug Abuse (NIDA), NIDA Office of Acquisitions – National Center for Advancing Translational Sciences (NCATS) Section, on behalf of the NCATS intends to negotiate and award a purchase order without providing for full and open competition (Including brand-name) to Sophion Bioscience.

(iii) The solicitation document and incorporated provisions and clauses are those in effect through Federal Acquisition Circular 2020-07, dated 08/03/2020.

(iv) The associated NAICS code 541990 and the small business size standard is $16.5M. This requirement has no set-aside restrictions.

(v) Project requirements:

The Stem Cell Translation Laboratory (SCTL) within the National Center for Advancing Translational Sciences (NCATS) Division of Preclinical Innovation (DPI) requires the service of Sophion to do automated whole-cell patch clamp (APC) using Qube instrument on iPSC derived sensory neuron in order to functionally characterize iPSC derived neuron for pain related screening projects. The services offered that are of interest to the SCTL include functional characterization of two ion channels (TRPV1 and P2X3) expressed in SCTL provide sensory neurons and separate into three projects:

1. Characterize VR1 (TRPV1) receptor expressed in iPS neurons

a. Calculate percentage of neurons that express sufficient VR1 current.

b. Carry out CRC for VR1 agonist: Capsaicin.

c. Carry out CRC for VR1 antagonist: Capsazepine.

2. Characterize P2x3 receptor expressed in iPS neurons

a. Calculate percentage of neurons that express sufficient P2x3 current.

b. Carry out concentration response curve (CRC) for P2x3 agonist: -MeATP

c. Carry out CRC for P2x3 antagonist: TNP-ATP.

3. Characterize cross talk between P2x3 and VR1 (TRPV1) receptors expressed in iPS neurons

a. Calculate percentage of neurons that express both P2x3 and VR1 current.

b. Carry out the current interaction when both receptors are activated comparing to when only single one is activated

These services will be utilized by the SCTL to obtain preliminary data in order to study the electrophysiology of voltage-gated ion channels of human pluripotent stem cells and their differentiated progeny, with a particular focus on sensory neurons. The data obtained from this study will be used to assess the functionality and heterogeneity of iPSC derived neuron. The long term goal is to applied this technology as a functionality screening platform for iPSC derived neurons.

(vi) Delivery shall be to:

The National Center for Advancing Translational Sciences (NCATS)

9800 Medical Center Drive

Rockville, MD 20850

One-time delivery to ship within 30 days ARO (vii) The provision at FAR clause 52.212-1, Instructions to Offerors – Commercial Items, applies to this acquisition.
