Statement of Work Summary: 75N98026Q00996
This is a Statement of Work (SOW) for a Firm Fixed-Price Purchase Order issued by the National Cancer Institute (NCI) under the National Institutes of Health (NIH) for the production of ChAdOx-1 and MVA viral vectors expressing mouse gp70. The work supports development of a viral vector-based cancer vaccine to evaluate efficacy in preventing brain metastases of breast cancer and treating glioblastoma.
The contractor must complete three primary technical tasks: (1) design and construct a codon-optimized gene expression cassette encoding mouse gp70 and mouse class II major histocompatibility complex invariant chain (Ii) suitable for expression in mouse cells and compatible with viral vector systems; (2) produce a ChAdOx-1 viral vector encoding the gene construct suitable for in vivo experimental use; and (3) produce an MVA viral vector encoding the same gene construct for in vivo experimental use. The contractor must also develop a project management plan including overall project timeline and key milestones. Minimum qualifications required include demonstrated experience designing codon-optimized gene constructs for viral vector applications, proven capability to produce both ChAdOx-1 and MVA viral vectors, and ability to perform work in an efficient, reliable, and professional manner. The period of performance is six months from award date, with all work performed at the contractor's facility. Two deliverables are required: completion of viral vector production due three months from award and a final report due at the end of the performance period, both submitted electronically to the COR/TPOC. Payment is contingent upon successful submission and acceptance of deliverables, with invoices submitted following COR approval. The Contracting Officer's Representative is Alan Peigh at NCI/NIH, and the Technical Point of Contact is Sharon Bargo at the Laboratory of Integrative Cancer Immunology.

75N98026Q00996 - STATEMENT OF WORK
1.0 TITLE
Production of ChAdOx-1 and MVA expressing mouse gp70
2.0 BACKGROUND
Glioblastoma, a high-grade primary brain tumor, is one of the most aggressive and devastating cancers. Despite intensive treatment, including surgical resection, chemotherapy, and radiation therapy, the median survival remains less than 14 months, and the 5-year survival rate is below 10%.
There is a critical need for new therapeutic approaches.
Immunotherapy has transformed cancer treatment and has been approved for more than 30 cancer types, particularly for patients whose disease is refractory to conventional therapies. However, effective immunotherapies for primary brain tumors, including glioblastoma, remain limited.
We have developed a viral vector-based cancer vaccine using a combination of chimpanzee adenovirus (ChAdOx-1) and modified vaccinia Ankara (MVA) vectors in syngeneic orthotopic mouse glioblastoma models. This approach significantly prolonged survival in tumor-bearing animals by inducing a robust population of tumor antigen-specific CD8+ T cells in the brain. These T cells were sufficient to mediate the protective effects of the vaccine.
Based on these findings, we will evaluate the efficacy of this vaccine strategy in preventing brain metastases of breast cancer, an area of significant unmet clinical need. To support this work, it is necessary to develop viral vectors expressing the mouse tumor antigen gp70 in combination with the mouse class II major histocompatibility complex invariant chain (Ii).
3.0 OBJECTIVES
The primary objective of this project is to develop ChAdOx-1 and MVA viral vectors expressing the mouse tumor antigen gp70 in combination with the mouse class II major histocompatibility complex invariant chain (Ii).
4.0 SCOPE
The scope of this project includes the following:
1. Design and construction of a codon-optimized gene expression cassette encoding mouse gp70 and the mouse class II major histocompatibility complex invariant chain (Ii) for expression in mouse cells.
2. Production of a ChAdOx-1 viral vector encoding the gene construct.
3. Production of an MVA viral vector encoding the same gene construct.
4.1 TECHNICAL REQUIREMENTS
The Contractor shall perform the following tasks:
4.1.1 Project Management Plan
The Contractor shall develop and submit a project management plan that includes the overall project timeline, key milestones, and a description of the approach to complete all required tasks.
4.1.2 Gene Construct Design
The Contractor shall design and generate a codon-optimized gene construct encoding mouse gp70 and mouse class II MHC invariant chain (Ii), suitable for expression in mouse cells and compatible with viral vector systems.
4.1.3 ChAdOx-1 Vector Production
The Contractor shall produce a chimpanzee adenovirus (ChAdOx-1) viral vector encoding the gene construct. The vector shall be suitable for in vivo experimental use.
4.1.4 MVA Vector Production
The Contractor shall produce a modified vaccinia Ankara (MVA) viral vector encoding the same gene construct. The vector shall be suitable for in vivo experimental use.
4.3 PERSONNEL QUALIFICATIONS
The Contractor shall meet the following minimum qualifications:
• Demonstrated experience in designing codon-optimized gene constructs for viral vector applications
• Proven capability to produce chimpanzee adenovirus (ChAdOx-1) vectors
• Proven capability to produce modified vaccinia Ankara (MVA) viral vectors
• Ability to perform work in an efficient, reliable, and professional manner
